Item ASSESSMENT OF FECUNDITY, HATCHING & SURVIVAL RATE OF NILE TILAPIA (Oreochromis niloticus) APPLYING FEED WITH GARLIC AND GINGER EXTRACT(DEPARTMENT OF BIOTECHNOLOGY, SHER-E-BANGLA AGRICULTURAL UNIVERSITY, DHAKA-1207, 2021-12) NAYEEM, DIDER HAIDERThe study was conducted at commercial hatchery at BRAC Tilapia Hatchery, Magura, Bangladesh and the analytical studies were conducted at Aquaculture Laboratory, Sher-e-Bangla Agricultural University to evaluate the effect of Garlic and Ginger extract in Nile tilapia diets on their reproductive performance (fecundity and hatching), survival rate on produced fry and hematological parameters of fry. A treatment was used as control without any extract. Nile tilapia (O. niloticus) with average weight of 267 g ± 3.16 were stocked for spawning into nine hapas at a density of 13 fish (10 females and 3 males) per 10 sq meter hapa, Three hapas with same diameters were used as replicates for each treatment. Hapas were fixed in an earthen pond (5 decimals). Feeding was done at a rate of 0.5% from their total biomass. Water quality parameters (temperature, pH, dissolved oxygen, ammonia) were measured weekly. Production of seed (eggs and fry) was compared with the tested diets. The highest mean of egg number (132.05 pcs) & mean spawn number (98.29 ± 1.11 pcs) per gram were obtained with the diets contained 0.02% Garlic extract (Treatment 2). Survival rate (84.24±0.05%) of fry was also found best with Treatment 2. Tilapia diets supplemented with Garlic extract had best responded on their reproductive performance (Hatching 80.63% ±1.05) and highest value FCR (1.43±0.19) & SGR (1.50 ± .19). The garlic extract treatment showed excellent performance for all the parameter under studied. Hence, the feeding with (0.02%) garlic extract can be used for large scale in tilapia breeding.Item ASSESSMENT OF HERBAL EXTRACT FOR GROWTH AND NUTRITIONAL QUALITY OF JUVENILE FRESHWATER PRAWN (Macrobrachium rosenbergii) UNDER BIOFLOC SYSTEM(DEPARTMENT OF BIOTECHNOLOGY, 2020) ISLAM, MD. SYFULThe study was conducted in the Biofloc Lab, Department of Aquaculture, Sher-eBangla Agricultural University to assess the potentiality of Macrobrachium rosenbergii, growth performance and its nutritional quality in Biofloc Technology (BFT) from July to November, 2020. Total five treatments were applied including T1 (Control), T-2 (Extract of Ginger), T-3 (Extract of Amla), T-4 (Extract of Garlic) and T-5 (Extract of Garlic and higher stocking) with three replications. The initial mean stock weight of prawn were 3.14±0.19, 3.04±0.27, 3.26±0.16, 3.36±0.06 and 3.02±0.36g in T-1, T-2, T-3, T-4 and T-5 respectively. After completing the research duration, the highest mean weight gain and percentage weight gain were 10.9±0.3g and 297.8±16.2% noticed in the T-3. The survival rate was maximum (99.5±0.8%) in the control treatment. The best FCR and SGR were 1.2±0.1 and 2.1% also noticed in the T-3. During the research 35% protein containing commercial feed was given and floc was produced at balanced level which was found 21.7% protein. From the proximate composition analysis, the highest protein percentage of prawn 74.13±.68% was found in the T-3. The best growth performance and nutritional value was observed in T-3 (Extract of Amla) compared to other treatments and also tells the immense possibility of M. rosenbergii culture in biofloc in the era of aquaculture and microbial biotechnology for mitigating the demand of animal protein.Item ASSESSMENT OF STRESS-TOLERANCE ATTRIBUTES IN WHEAT USING GENE – SPECIFIC MOLECULAR MARKERS(Department of Biotechnology, 2014-12) MAHMUD, RONAItem BIOFLOC CULTURE METHOD FOR GROWTH PERFORMANCE, SURVIVAL RATE AND NUTRITIONAL COMPOSITION OF SNAKEHEAD FISH (Channa striata) ON NATURAL EXTRACT SUPPLEMENT(DEPARTMENT OF BIOTECHNOLOGY, 2020) ALAM, MD. KHAIRULSnakehead fish (Channa striata) is a well known and important freshwater fish in Southeast Asian countries. The experiment was conducted at Sher-e-Bangla Agricultural University Biofloc Culture Lab to evaluate the growth performances, survival rate and nutritional composition of snakehead fish (C. striata). There has four treatments with three replications. Water quality parameters were monitored through temperature, dissolved oxygen, pH, ammonia, total dissolved solids, alkalinity and hardness. Maximum length increase (14.15±0.2) cm of C. striata was found in treatment-4. Highest weight gain also noticed in the same treatment which was (105.67±6.65)g. Control treatment showed lowest performance among the parameter studied. Maximum specific growth rate was 2.70% in treatment-4 whereas 2.50% was minimum in treatment-1. In treatment-3 survival rate was highest, which was 75.76% and in control treatment, the lowest survival rate was recorded that was 47.88%. Feed conversion ratio was better (1.08) in tretment-4. Daily mean weight gain was noteworthy in treatment-4 which was 1.50g and lowest daily mean weight gain was recorded 1.1g in control treatment. From the analysis of proximate composition of C. striata 69.67% protein content was available in treatment-4 which was maximum and the lowest protein content was found 61.84% in treatment-2. Considering all the results, the optimum growth performance and nutritional composition was better in treatment-4 (Extract of Garlic and Amla) compared to other treatments. In the era of aquaculture and microbial biotechnology there has a huge potentiality to fulfill the requirement of fish protein by the culture of C. striata in biofloc technology.Item BIOINFORMATIC ANALYSIS OF MITOCHONDRIAL GENOME OF HUMPED FEATHERBACK CHITAL FISH, Chitala chitala (HAMILTON, 1822)(DEPARTMENT OF BIOTECHNOLOGY, SHER-E-BANGLA AGRICULTURAL UNIVERSITY, DHAKA-1207, 2021-12) IRTIZA, MD. HASINThe present study was about the bioinformatic analysis of mitochondrial genome of an endangered featherback fish, Chitala chitala. Although Chitala chitala plays significant role in inland fishery, but its’ evolution has received little attention. In the current study, the mitogenome of Chitala chitala was retrieved and investigated. Some of the bioinformatics tools were applied to measure important gene parameters including AT/GC-skewness, codon adaptation index (CAI), the effective number of codons (ENc) and GC percentages of each protein coding gene. The majority of AT-skew and GC-skew values of all protein coding genes of the species were negative, and the amplitude of the GC-skew was larger than the AT-skew. On the basis of comparative selective strength analysis with the PCGs of two related mitogenomes, I discovered that most of the PCGs retained the Ka/Ks ratios less than one implying that they evolved under strong negative or purifying selection. Moreover, I analyzed the codon frequency and relative synonymous codon usage (RSCU). Then, I identified a total of 22 RSCU values (>1) and revealed 6 codons as “over-presented” that implied for codon usage bias to engage in highly expressed genes for efficient protein synthesis via translational selection. The phylogenetic tree and time tree were constructed by Maximum Likelihood (ML) method, providing further supplement to the evolution of the fish. Regarding on the facts of the presence of codon usage biasness rolling in translational selection and the signs of purifying selection identified in PCGs indicate obvious conservation of this endangered fish species.Item CALLUS INDUCTION AND MICROPROPAGATION OF PINEAPPLE (Ananus comocus L.)(Department of Biotechnology, Sher-e-Bangla Agricultural University, 2015-12) AKTER, FATEMAThe present study was undertaken in Biotechnology Laboratory of the Department of Biotechnology, Sher-e-Bangla Agricultural University, Sher-e-Bangla Nagar, Dhaka 1207 during the period from January. 2014 to December. 2014 to evaluate the effect of different plant growth regulators on callus induction, shoot proliferation, root formation and to develop micropropagation protocol of pineapple. Crown tip of pineapple fruits were used as explants and inoculated in MS media supplemented with different cone. of Benzyl adenine (BA), Kinetine (KIN). Indolebutyric acid (IBA). Naphthaleneaeetie acid (NAA) alone or in combination with each other. The treatment 3.0 mgIL KIN + 2.5 mg/L NAA showed the highest percentage (800/0) of callus induction. But minimum days (21.20) to callus induction and maximum weight of callus (2.80 gm) was observed with 2.0 mg/L KIN + 1.5 mg/L NAA treatment. The highest 85% shoot induction was obtained with 3.0 mgfL KIN +1.0 mg/L NAA treatment and minimum 7.60 days was recorded with the same treatment. The highest number of shoots (18.40) after 6 weeks of subculture was recorded in 3.0 mg/L KIN + 1.0 mg/L IBA and the longest shoot (9.04 cm) was found with 3.0 mg/L KIN+2.0 mg/L IBA. The treatment 2.0 mg/L IBA+ 1.5 mg/L NAA showed the highest percentage (75%) of root induction, minimum 16 days to root induction and the longest root (7 cm). But the highest number of roots (15.60) per explants was recorded with 3.0 mg/L IBA+ 1.0 mg/L NAA after 6 weeks of subculture to rooting media. Regenerated plantlets showed, 85% survival in growth chamber conditions and 82% in shade house stage of hardening with Sand : Soil : Cow-dung (1:1:1) and 78 % in open atmosphere at direct sunlight.Item CALLUS INDUCTION AND PLANT REGENERATION OF STEVIA (Stevia rehaudiana B.)(Department of Biotechnology, Sher-e-Bangla Agricultural University, 2015-12) FERDOUS, JANNATULThe present research work was carried out in Biotechnology laboratory of the Department of Biotechnology. Sher-e-Bangla Agricultural University. Shcr-e-Bangla Nagar, Dhaka- 1207 from the period of January 2014 to December 2014 to evaluate the effects of different plant growth regulators in in vitro on shoot proliferation and root lbrmation and finally to develop a viable micropropagation protocol in Stevia (Stevia rehaudiana) using shoot tip and nodal segments as explants. Explants was inoculated in MS (Murashige and Skoog) media supplemented with the Benzvladenine (BA) and Kinetine (KIN) for direct shoot formation. Combination of BA and Indolebutyricacid (IBA) as well as combination of KIN and IBA were used in MS medium for indirect shoot formation via callus induction. The highest percent of shoot induction (83.42%) obtained on 2.0 mg/L BA and the minimum days to shoot induction (10.6) were also obtained on 2.0 mg/I.. BA. The highest 10.8 shoots and 17.2 leaves per explants also observed in BA 2.Omg/L. But maximum average length of shoots per explants (3.94 cm) were reported from media containing 1mg/I. KIN. Highest callus induction potentiality (83.58%) was observed in 2 mg/L BA+1.5 mg/L IBA The minimum days to callus induction (13.4) were recorded in 1.0 mg/L KIN+I.5 mg/L IBA but the minimum days to shoot induction (9.6) from callus were recorded in 2.0 mg/L BA+1.5 mg/L IBA. Righest percent of root induction (78.18%) with 7.2 roots per explants and minimum 12.6 days to root induction were noticed on 1.0 mg/I. IBA. The combined ellèct on percent of' root induction was highest (76.92%) in the treatment of 1.0 mg/L KIN t 0.5 mg/L IBA. The same treatment showed minimum days (13.4) to root induction and maximum number of roots per explants. In acclimatization 90%, 80% and 80% survival rate were recorded in growth cabinet, shade house and field condition respectively. In vitro regeneration technique was found to be very effective and promising niethod in the proliferation of Stevia and this method can be used 11w large scale production of disease free and quality planting material.Item COMPARATIVE PERFORMANCE OF AMMONIUM NITRATE (NH ) AND OTHER NITROGENOUS SALTS FOR IN VITRO REGENERATION OF POTATO (Solanum tuberosum L.)(DEPARTMENT OF BIOTECHNOLOGY, 2020) WAKIL, MIRAJ AHMEDAn experiment was undertaken at the Biotechnology Laboratory, Department of Biotechnology, Sher-e-Bangla Agricultural University (SAU), Dhaka, Bangladesh during the period of January, 2018 to January, 2019 to identify a new chemical for the substitute of explosive chemical Ammonium nitrate (NH 4 NO ) in MS media composition for in vitro regeneration of potato (Solanum tuberosum L.). One potato variety Asterix was used as an experimental materials. The new chemical was denoted as α-chemical for the privacy of the experimental findings. MS powder, MS liquid standard dose, different concentrations (1, 5, and 10 gmL 3 ) of α chemical which have twice dose of other component in stock solution-I for the preparation of tissue culture media were used as treatment. It was noticed that in vitro regeneration of potato has been successfully done with α-chemical in new composition of tissue culture media. The shoot length, node number, number of leaf, root length were highest in MS powder (T ) at 14, 21 and 28 days after inoculation (DAI). Although, in some parameters MS powder (T ) gave maximum result but it was statistically non-significant with 5gm of α- chemical/litre in stock solution-I having twice dose of other components in new media composition. The treatment 5gm of α-chemical/litre in stock solution- I having twice dose of other component showed average performance in all the traits under studied. MS powder, Liquid MS standard dose and 1 gmL 0 -1 in stock solution- I twice dose of α chemical showed comparatively similar result on most of the parameters under studied. The newly used α-chemical is cheap, nonexplosive and environmental friendly which can be an alternative of destructive chemical Ammonium nitrate (NH 4 NO ) for tissue culture media preparation.Item DNA BARCODING OF SILURIFORM FISHES OF THE MEGHNA RIVER ESTRUARY(DEPARTMENT OF BIOTECHNOLOGY, SHER-E-BANGLA AGRICULTURAL UNIVERSITY, DHAKA-1207, 2021-12) MOLLIK, NIAZ MORSHEDThe Meghna river estuary is the largest estuarine ecosystem in Bangladesh and the fish assemblage in this estuary is highly diverse. Estuaries are also very important as breeding and nursery grounds for a wide variety of fishes. Fish samples were collected from the different zone of the Meghna estuary around the year July 2020 to May 2021. In the present study, a total of 9 species were identified from the 7 seven genera and two families (Bagridae and Schilbeidae) under the order Siluriformes based on DNA barcoding approach along with morphology. The species were M. bleekeri, M. gulio, M. vittatus, R. rita, S. aor from the Bagridae family and A. coila, C. garua, E. vacha, S. silondia from the Schilbeidae family. The barcode sequences obtained from the present study showed high identity (> 99%) with the reference sequences deposited in the GenBank database. The assessment of species identities with previously known sequences and closely related species in NCBI BLAST generated 99% of identities indicating the effectiveness of COI sequences to provide species-level resolution. Sequences of the present study were clustered in a single clade with the sequences of conspecific species from GenBank which validate the genetic identification of the studied species. Genetic distances among the sequences of the present study and its compared sequences from GenBank ranged from 0.00 (0.2%) to 0.003 (0.3%) which also validates the genetic identification of this species. Fish diversity in the Meghna has long been declining due to the number of anthropogenic and natural factors. This study provides information on the species diversity of Siluriformes that will help in the management and conservation of the fish fauna of the Meghna river system.Item DNA FINGERPRINTING AND MOLECULAR DIVERSITY ANALYSIS OF GINGER (Zingiber Officinale) GENOTYPES USING RAPD MARKERS(Department of Biotechnology, 2017) FATEMA- TUZ- ZOHURAGinger (Zingiber officinale) rhizomes have been widely used as a spice and flavoring agent in foods and beverages in Bangladesh as well as in all over the world for its economical and medicinal values. Identification and characterization of germplasm is an important link between the conservation and utilization of plant genetic resources. The present investigation was undertaken for the assessment of 13 local genotypes of Zingiber officinale collected from different region of Bangladesh by 7 RAPD random decamer primers. Good quality genomic DNA was extracted from ginger genotypes using CTAB method. Nine RAPD primers were screened for amplification of genomic DNA and seven primers were selected based on the amplification pattern. A total of 34 distinct and differential amplification bands ranging from 150-1200 kb were observed with an average of 1.14 polymorphic bands per primer. The overall gene diversity was detected 0.8052 and the value of PIC was detected 0.7532. Primers could generate enough polymorphism for possible use in diversity studies, based on provisional multivariate analyses such as cluster analysis and principal component analysis (PCA). PCA classified 13 ginger genotypes into four groups and showed in two dimensional scatter plot. The genetic similarity coefficients among genotypes ranged from 0.103 to 0.654. Cluster analysis based on Jaccard’s similarity-coefficient using UPGMA grouped the genotypes into two clusters: Cluster A and Cluster B. The cluster ‘A’ had only one genotype Kaptai local and the second cluster ‘B’ had rest of twelve genotypes. Further cluster B subdivided into another cluster. The prevalence level of polymorphism in the local genotypes of ginger will help to breeders for ginger development breeding program.Item DNA FINGERPRINTING AND MOLECULAR DIVERSITY OF MUNG BEAN (Vigna radiata (L.) R. Wilczek) GERMPLASM(DEPARTMENT OF BIOTECHNOLOGY, 2022) TAWFIQ, TAMANNAMung bean (Vigna radiata (L.) R. Wilczek) has been widely used as a pulse crop. The study was carried out at the Biotechnology laboratory of Sher-e-Bangla Agricultural University, during the period of July 2021 to June 2022 to investigate molecular diversity and fingerprinting of fifteen mung bean germplasm. RAPD primers were selected and synthesized for molecular diversity analysis. All of the primers were able to produce polymorphic bands. The highest number of bands were observed in RAPD primer OPT20. The polymorphic Information Content (PIC) of RAPD markers ranged from 0.24 to 0.32 with an average value of PIC =0.27. Gene diversity ranges from 0.41 to 0.28 with an average value of 0.33. Primer showed OPBC-1, OPA-20, OPP-03, OPT-20 moderate diversity and primer OPBC 18 showed low diversity on the basis of PIC value. The value of pair -wise comparisons of Nei’s (1983) genetic distance (D) among 15 genotypes was computed from and it was ranged from 0.1034 to 0.6207 with an average of 0.3621. Dendrogram based on Nei’s genetic distance using unweighted Pair Group Method Of Arithmetic Mean (UPGMA) indicated the segregation of 15 genotypes into four main clusters. This result can be conveniently used for molecular diversity analysis to identify diverse parent for mung bean improvement program.Item DNA MARKER BASED HYBRIDITY TEST OF DIFFERENT EGGPLANT (Solanum melongena L.) HYBRIDS(Department of Biotechnology, 2017) HOSSAIN, MAHBUBAAn experiment was carried out to identify the hybrid variety of eggplant through SSR marker. Four eggplant hybrids vizMEH5003, MEH5006, MEH5009, MEH5012 and their corresponding parents were used as experimental materials. The hybrid varieties of eggplant were developed by Metal Seed Ltd., Dhaka, Bangladesh. The experiment was conducted at the Department of Biotechnology, Sher-e-Bangla Agricultural University, Dhaka, Bangladesh. Initially different SSR primerswere used to establish polymorphism among the parents of each hybrid. The identified polymorphic primers were used to screen the hybrid variety together with corresponding parents. It was noticed that only one primer viz MEH SSR-04 showed polymorphism in the parents of MEH5009 hybrid. This primer was used for the detection of hybrid variety. A 150bp DNA fragment was amplified in male parent and hybrid variety which was absent in female genotype. This DNA band can be used for large scale screening of that specific hybrid variety for seed purity in commercial purpose. The parents of other hybrid showed monomorphic DNA band and some of the SSR primerswere not amplified in any eggplant genotype under investigation. The confirmation of genetic purity in seeds is very important for identification of genetic contamination and hybrid purity for farmers and plant breeders before sale of the seed to public domain. CULTIVARS USING RAPD MARKERS(Department of Biotechnology ,Sher-e-Bangla Agricultural University , Dhaka 1207, 2014-12) SULTANA, SHARMINAn experiment was conducted on Brinjal (Solanum melongena L.) in Biotechnology Laboratory of the Department of Biotechnology, Sher-e-Bangla Agricultural University, Dhaka-1207. The genetic variation of 11 brinjal varieties (BARI begun 1, BARI begun 4, BARI begun 5, BARI begun 6, BARI begun 7, BARI begun 8, BARI begun 9, BARI begun 10, BARI hybrid begun 1, BARI hybrid begun 3 and BARI hybrid begun 4) were studied based on PCR technology using five RAPD primers. Sum total of 44 distinct DNA amplified band were observed with an average of 5.6 polymorphic bands per primer. The overall gene diversity was detected 0.2186 and level of polymorphism was detected 63.64%. The pair wise inter-variety similarity indices ranged from 67.03% to 97.61%. The UPGMA Dendrogram segregated 11 genotypes of brinjal into two main clusters. The first major cluster had only one genotype (BARI begun 6) and the second major cluster had rest of ten genotypes. BARI begun 6 vs. BARI begun 1 showed highest Nei’s genetic distance (0.5261) as they are released from different parental origin. On the other hand BARI began 9 vs. BARI begun 7 varietal pair showed lowest genetic distance (0.0465) as they are released from same parental origin. The experiment reveals significant level of variations among 11 brinjal genotypes by RAPD markers. In pot culturecondition treatment 500 ppm C i.' ;respwidcd excellent in all the parameters under stud.Item EFFECTS OF DIETARY SUPPLEMENTATION OF Sargassum vulgare ON GROWTH AND HEMATOLOGICAL PARAMETERS OF ORNAMENTAL FISH KOI CARP (Cyprinus rubrofuscus)(DEPARTMENT OF BIOTECHNOLOGY, SHER-E-BANGLA AGRICULTURAL UNIVERSITY, DHAKA-1207, 2021-12) HOSSAN, MOHAMMAD IQBALA research was conducted to assess different formulated diets supplemented with different doses of seaweed (Sargassum vulgare) on ornamental fish Koi carp (Cyprinus rubrofuscus) in the aquaculture laboratory of Department of Aquaculture, Sher-e-Bangla Agricultural University to evaluate the growth and hematological parameters. Total four treatments were applied including Treatment- 1 (Control, 0% seaweed), Treatment-2 (5% seaweed), Treatment-3 (10% seaweed) and Treatment-4 (15% seaweed) each with three replicates. Four iso-proteinase feed was formulated where average percentage of protein, lipid, fiber and carbohydrate in formulated diet were 27.59%, 6.1%, 7.59 and 48.96%. The initial mean weight of fishes was 14.4±2.12, 14.5±0.75, 13.1±2.4 and 12.8±1.43 g in T-1, T2, T-3 and T4 respectively. The experiment was concluded in 100L glass aquarium with mechanized water filter system for each for 8 weeks and weekly sampling were done. Feed was applied twice a day. Temperature, pH, TDS, NH 3, NO 2 - ii and NO 3 - data were collected daily during the experimental period and there was no significant difference in water parameters among treatments. Mean weight (g) of fish, length (cm) and width (cm) data were collected weekly basis. The mean weight gain was found 7.13±1.84, 8.06±1.58, 6.98±1.84 and 4.72±0.52 in T-1, T-2, T-3 and T-4 respectively. The highest weight gain was found in T-2 (5% seaweed) and the percentage of weight gain 55.86% found in the same treatment. There was no mortality found in T-2 and T-4. Survival rate were found 96.6% in Control. The lowest FCR 1.43±0.26 found in T-2 and SGR also found highest in the same treatment. Hematological parameter showed that blood hemoglobin and glucose level was higher in T-2 than others treatments which were 11.6±0.17gm/dl and 6.33±0.26 mmol/l. Serum protein was found 6gm/dl in T-2 and lowest value was found in control. Spleen Somatic Index (SSI) percentage of experimental fishes was also found higher in T-2 and lowest one was recorded in T-1. Refractive index (RI) of blood serum was found better in T-2 fishes compared to other treatments. It can be concluded that feed supplemented with 5% seaweed (T-2) diet showed better growth performance and hematological parameters compared to other treatments. Feed supplemented with brown seaweed Sargassum vulgare as a dietary ingredient can be used for better immunity and growth performance of ornamental fish.Item GENETIC DIVERSITY ANALYSIS OF DEEP WATER RICE AND SCREENING OF RICE (Oryza sativa L.) GENOTYPES FOR BACTERIAL LEAF BLIGHT(DEPARTMENT OF BIOTECHNOLOGY, 2022) JANNAT, KAZI MEFTAHULGenetic diversity analysis was performed to investigate polymorphism among deep water rice and molecular screening to identify bacterial leaf blight disease resistant gene containing rice genotypes. In the flooding region, most of the deep water rice genotypes is drown due to flood. Besides, rice genotype also affected by bacterial leaf blight disease which also causes vast yield loss. The experiment was carried out with 15 and 41 rice genotypes with five well known SSR and five gene specific markers amplification respectively. In case of 15 deep water rice, fifty nine alleles ranged from 7 to 16 alleles per locus and an average of 11.8 was detected. Forty seven polymorphic alleles were also identified. The Polymorphic Information Content (PIC) of SSR markers ranged from 0.103 (RM206) to 0.494 (RM252) with an average value of PIC 0.317. Three primers showed moderate diversity and two primers showed low diversity on the basis of PIC value. The RM252 marker can be considered as the best marker for giving highest number of band. The similarity among rice genotypes was computed from combined data for the 5 primers that ranged from 0.3729 to 0.9375 with an average of 0.7048. Dendrogram indicated the genetic distance of 15 deep water rice genotypes by grouping them into four main clusters. In case of gene specific markers Xa4, xa5, Xa7, x13 and Xa21 bacterial leaf blight disease resistant genotypes were identified carrying bacterial leaf blight resistant genes. Among 41 rice genotypes, two different genes were present in 10 genotypes (Dudhsor, Dula Bech, Dulai Aman, Kata Mukul, Anguli Aman, Katari, BR16, BR18, BRRI dhan29 and BRRI dhan58) and one separate gene was present in 12 genotypes. Phylogenetic tree represents the genetic relatedness of 41 rice genotypes by diving them into two main clusters with sub clusters. These results can be conveniently used for identifying diverse parent for the development of high qualitative and quantitative characteristics bearing genotypes in rice.Item IDENTIFICATION OF A NEW CHEMICAL AS A SUBSTITUTE OF AMMONIUM NITRATE (NH 4 NO ) IN MS MEDIA PREPARATION FOR IN VITRO REGENERATION OF POTATO (Solanum tuberosum L.)(Department of Biotechnology, Sher-e-Bangla Agricultural University, 2016-12) BASHAR, MD. ABULAn experiment was undertaken at the Biotechnology Laboratory, Department of Biotechnology, Sher-e-Bangla Agricultural University (SAU), Dhaka, Bangladesh during the period of January, 2015 to June, 2016 to identify a new chemical for the substitute of explosive chemical Ammonium nitrate (NH 4 NO ) in MS media composition for in vitro regeneration of potato (Solanum tuberosum L.). Three potato varieties viz Diamant, Cardinal and Asterix were used as an experimental materials. The new chemical was denoted as β chemical for the privacy of the experimental findings. Different concentrations (1, 5. 10, 15 and 20 gmL 3 -1 ) of β chemical were used for the preparation of stock solution- A, which was finally used for MS media preparation. It was noticed that in vitro regeneration of potato has been successfully done with new composition of β chemical in MS media preparation. The shoot length, node number, number of leaf, root length were highest in 5 gmL -1 of β chemical at 14, 21 and 30 days after sub-culture (DAS). Although, in some parameters liquid MS standard dose (16.5 gmL ) gave maximum result but it was statistically non significant with 5 gmL of β chemical. All the plantlets were died at higher concentration (15 gmL and 20 gmL -1 ) of β chemical. It may be due to toxic effect of the chemical. The variety Diamant showed best performance in all the traits under studied. It is reveal that ready made MS powder, Liquid MS standard dose and 1 gmL of β chemical showed comparatively similar result on most of the parameters under studied. Among the varieties, the Asterix variety showed minimal performance in most of the phenotypic characters under studied. In all the case, Diamant variety showed best performance for all the phenotypic characters under studied. The newly used β chemical is cheap, non explosive and environmental friendly which can be an alternative of destructive chemical Ammonium nitrate -1 -1 -1 -1 (NH 4 NO ) for MS media preparation. 3Item IN VITRO PLANT REGENERATION OF Aloe vera L.(Department of Biotechnology, Sher-e-Bangla Agricultural University, 2014-07) ALAUDDIN, MD.The present research was carried out on Aloe vera L. in Biotechnology Laboratory of the Department of Biotechnology, Sher-e-Bangla Agricultural University, Sher-e- Bangla Nagar, Dhaka-1207 during the period from September, 2013 to July, 2014 to evaluate the effect of different plant growth regulators on multiple shoot proliferation and root formation along with acclimatization for in vivo survival and finally to develop a viable micropropagation protocol. Shoot tip of young lateral shoots (sucker) were used as the explants, which were sterilized using freshly prepared 0.1% HgCl2 solution with few drops of Tween-20. After sterilization, explants were inoculated in MS media supplemented with either singly or in combination of the Benzylamino purine (BAP), Kinetine (KIN), Indolebutyric acid (IBA), Naphthaleneacetic acid (NAA). The highest percentage of shoot proliferation (85%) and minimum days to shoot induction (13.6 days) were achieved on MS medium containing 1.0 mg/L BAP + 0.5 mg/L IBA. The highest 14.8 shoots and 24.4 leaves per explant were also observed in the same media composition. But the highest average length of shoots (9.62 cm) was recorded from media containing 1.5 mg/L BAP + 1.0 mg/L NAA. The highest percentage (95%) of rooting was obtained in media treated with 3.0 mg/L IBA along with maximum 12.4 roots per explant. But the highest (10.24 cm) average length of root was observed in 2.0 mg/L IBA. The highest (5.374 mm) diameter of root and minimum days to root induction (10.6 days) were noticed on the same culture media supplemented with 2.5 mg/L NAA. In regenerated plantlets, 100% survival in growth chamber conditions and 88% survival under shade house and 90% survival in open atmosphere were achieved. Micropropagation was found to be very effective and promising method in the propagation of Aloe vera and this protocol can be a useful tool for proliferation of Aloe vera.Item IN VITRO PLANTLET REGENERATION IN MINT (Mentha spicata)(DEPARTMENT OF BIOTECHNOLOGY, SHER-E-BANGLA AGRICULTURAL UNIVERSITY, DHAKA-1207, 2018-06) AHMED, SAYED SHAQURThe present study was undertaken in Biotechnology Laboratory of the Department of Biotechnology, Sher-e-Bangla Agricultural University, Sher-eBangla Nagar, Dhaka-1207, during the period from June, 2017 to January, 2018 to evaluate the effect of different plant growth regulators (BA, IBA) on in vitro plantlet regeneration in Mint. The shoot tips of Mint were used as explants which were inoculated in MS media supplemented with different conc. (1.0, 1.5, 2.0 and 2.5 mg/l) of Benzyl adenine (BA) and Indole-3-butyric acid (IBA) (0.5, 1.0, 1.5 and 2.0 mg/l) either alone or in combination of both. The experiments were arranged in Completely Randomized Design (CRD) with three replications. The maximum shoot induction was obtained with 2.0 mg/L BA. The highest shoot length (12.12 cm) was found in 2.0 mg/L BA and minimum shoot length (2.90 cm) in control treatment. The highest percentage (85.00%) of root induction was recorded with BA 2.0 mg/l + IBA 1.0 mg/l in minimum (11.0 days). The highest length of root (3.25, 5.00 and 7.50 cm) at 21, 28 and 42 DAI respectively was found in 1.0 mg/l IBA and the control treatment found the lowest length of root at all DAI. The treatment BA 2.0 mg/l + IBA 1.0 mg/l gave the highest number of root (10, 15 and 20.67) at 21, 28 and 42 DAI respectively. Regenerated plantlets showed 90% survival during in growth chamber and 83.33% in shade house and 80 % in open atmosphere at direct sunlight. Therefore, an efficient protocol has been developed for in vitro regeneration of Mint which has great commercial value for year round production in Bangladesh.Item IN VITRO PLANTLET REGENERATION OF CHRYSANTHEMUM (Chrysanthemum morifolium)(Department of Biotechnology, 2017) KHAN, MD. SHAHIDUL ISLAMThe present research was carried out in Biotechnology Laboratory of the Department of Biotechnology, Sher-e-Bangla Agricultural University, Dhaka-1207 from the period of February 2016 to June 2017 for in vitro regeneration of chrysanthemum where nodal segments were used as explants. Five levels of BA (1.0, 2.0, 3.0, 4.0 and 5.0 mg/l) and IAA (0.5, 1.0, 1.5, 2.0 and 2.5mg/l) were used for shoot and root induction. 2,4-D (0.5, 1.0, 1.5 and 2.0 mg/l) was used in combination with 2.0 mg/l of BA for callus and shoot induction. The highest frequency of shoot (80.00%) in minimum 7.60 days and maximum number of shoot (2.20, 2.60 and 3.00) at 14, 21 and 28 DAI (days after induction) were recorded from BA 2.0 mg/l. Moreover, the highest length of shoot and number of leaves was noticed from the 3.0 mg/l and 2.0 mg/l of BA, respectively at 14, 21 and 28 DAI. The highest frequency of callus (76.00%) was observed within 14.20 days in MS medium and weight of callus (1.80 g, 2.70 g and 3.50 g) was obtained with BA 2.0 mg/l+ 2,4-D 1.0 mg/l. The maximum number of shoot (3.20) at 40 DAI was obtained with BA 2.0 mg/l+ 2,4-D 1.0 mg/l within minimum 9.20 days. IAA 0.5 mg/l produced the highest percentage of root (80.00%) within 13.00 days while longer days required in other treatments. The maximum number of root (2.60, 3.60 and 5.40) was obtained from 1.5 mg/l IAA at 14, 21 and 28 DAI, respectively. The maximum length of shoot (2.48 cm, 2.84 cm and 3.66 cm) at 14, 21 and 28 DAI was noticed from the BA 2.0 mg/l+ IAA 1.0 mg/l. In combined effect the highest percentage (76%) of root induction was recorded with BA 3.0 mg/l+ IAA 1.5 mg/l in minimum 12.20 days and BA 2.0 mg/l+ IAA 1.0 mg/l gave the highest number of root 2.40, 3.20 and 4.20 at 14, 21 and 28 DAI, respectively. The survival rate was 80% in shade condition which was 75% in open atmospheric condition. Therefore, an efficient protocol has been developed for in vitro regeneration of chrysanthemum which has great commercial value for year round production in Bangladesh.
