Abstract
An experiment was jointly accomplished at Sher-e-Bangla Agricultural University and
ASRBC, ACI, Dhaka, Bangladesh to develop a protocol for in vitro regeneration of
Lisianthus during the period from January, 2015 to September, 2016. Response of leaf
explants different hormonal concentrations in different media were observed. Initiation
of callus and response towards shooting were earlier in MS medium than B5 medium
with 1.5 mg/l NAA supplementation. In case of shoot regeneration, four combinations
of BAP and GA
3
(0.5X1.0, 1.0X0.5, 1.5X1.0, 1.0X1.5 mg/l respectively) were used.
Highest mean number of shoot (16.3) and shoot length (2.6cm) were recorded when
supplemented with 1 mg/l BAP and 1.5 mg/l of GA
3
on MS medium. Full strength MS,
half strength MS and B5 media supplemented with different concentrations of IBA and
IAA were used for root initiation, where 1.5 mg/l of IBA was found to be the best for
rooting. In vitro regenerated plantlets were acclimatized in sand, vermicompost and
coco dust with the ratio of 1:1:1.
