Item VARIABILITY CORRELATION AND PATH ANALYSIS IN SEGREGATING POPULATION OF Brassica rapa OBTAINED THROUGH INTERVARITAL CROSSES(DEPARTMENT OF GENETICS AND PLANT BREEDING, SHER-E-BANGLA AGRICULTURAL UNIVERSITY, DHAKA, BANGLADESH, 2010-06) ARA, SHIFATA research was conducted by using eight F2& eight F4 populations generated through inter-varietal crosses, along with three check varity of Brassica rapa and grown in the experimental farm of Sher-c-Bangla Agricultural Unriversity. Dhaka, during November 2009-March 20010 to study the variation in different characters, correlation between pairs of different characters and the direct and indirect effect of different characters on seed yield per plant of the F2 & F4 materials to select the plants with higher potential. From the values of mean, range and CV (%) of seed yield and yield contributing characters it was confirmed that there were considerable variation present among all the genotypes used in the experiment. It was shown that more seggregation present in P2 material than F4 material. The values of phenotypic variances were higher than the corresponding genotypic variances. I)ays to 50% flowering, days to maturity, number of primary branches per plant, number of secondary branches per plant, length of siliqua, number of seeds per siliqua. thousand seed weight and yield per plant showed least difference between phenotypic and genotypic variances. The values of GCV and PCV indicated that there was least variation present among most of the characters. The days to maturity, length of siliqua, seeds per siliqua, thousand seed weight showed high heritability with low genetic advance and genetic advance in percentage of mean. Yield per plant had significant and highest positive correlation with length of siliqua, seeds per siliqua and thousand seed weight. The path co-efficient analysis revealed that siliquae per plant had the highest positive direct eRect followed by number of secondary branches per plant, days to 50% flowering, length of siliqua, and plant height. Dhaka. during April 2009 to September 2010. The objectives of the study were to measure the variability among the genotypes for yield and yield contributing characters, estimate genetic parameters, association among the characters and their contribution to yield. There was a great deal of significant variation for all the characters among the genotypes. Considering genetic parameters high genotypic co-efficient of variation (GCV) was observed for branches per vine, yield per plant and number of fruit per plant whereas low genotypic co-efficient of variation (GCV) was observed for days to first male and female flowering. In all the cases, it was found that phenotypic coefficient of variation was greater than genotypic co-efficient of variation. }lighest genotypic and phenotypic co-efficient of variation was observed in branch per vine, fruit length, fruit weight and number of fruit plant which indicated a wide variability among the genotypes and offered better scope of selection. The results obtained showed that fruit length showed low direct and positive effect on yield per plant and indirect positive effect on yield per plant via fruit diameter and average fruit weight. Similar result was found for fruit diameter. Average fruit weight and number of fruits per plant showed high direct and positive effect on yield per plant. Path analysis revealed that average fruit weight, number of fruits per plant, days to male flowering and fruit length had positive direct effect on fruit yield. Hybridization between the genotypes of cluster II and cluster VI will manifest maximum heterosis and create wide genetic variability. The highest heterosis would be manifest in cross combination involving the genotypes belonging to divergent clusters. Considering group distance and the agronomie performance, the inter genotypic crosses between G2& 65; G2&G14;O I 4&G I 5:G2&G I 5;G I 0&G II ;G I 0&Gl 3:611 &Gl 3;G5&G I 5;G5&G I 4;might be suitable choice for future hybridization programme.Item COMBINING ABILTY AND HETEROSIS THROUGH LINE X TESTER ANALYSIS IN Brassica napus L.(DEPARTMENT OF GENETICS AND PLANT BREEDING, SHER-E-BANGLA AGRICULTURAL UNIVERSITY, DHAKA, BANGLADESH, 2010-06) HASSAN, OLIULThe study was conducted to evaluate six female parents (CMS lines) and 3 male parents (testers) in a line x tester mating design at the research field of Sher-e-Bangla Agricultural University, Dhaka, during November 2009 to March 2010 to estimate their combining and heterosis. The data recorded on 18 F1 s and their parents for their combining ability indicated that (ICA effect was significant for days to fifty percent flowering, plant height, No. of siliqua per plant, seed per siliqua, primary branch & secondary branch per plant and seed yield per plant . The specific combining ability (SCA) effect was significant for all the characters. High ratio of SCA and GCA variance was observed, indicating preponderance of non additive gene effects in the inheritance of the yield and yield relating characters under study. Among the lines, CMS2, CMS3 and CMS5 were found as good general combiners. But seven hybrids exhibited good specific combiner among eighteen hybrids. Different types heterosis i.e. heterosis over mid parent (Hm), heterosis over better parent (Rb) and heterosis over standard check (HC) were estimated to evaluate eighteen hybrids for seed yield and yield related characters, where BARI Sharisha-7 was taken as check. The average heterosis for seed yield of eighteen hybrids over mid parent was 19.26% and that of better parent and standard check was 7.96% and 73.90%, respectively. Dhaka, during October 2009 to March 2009. The objectives of the study were to measure the variability among the genotypes for yield and yield contributing characters, estimate genetic parameters, association among the characters and their contribution to yield. There was a great deal of significant variation for all the characters among the genotypes. High genotypic co-efficient of variation (CCV) was observed for pod width, inflorescence length, pod per inflorescence whereas low genotypic co-efficient of variation (('CV) was observed for seed Nvidth, seed length, days to first flowering. In all cases, phenotypic variances were higher than the genotypic variance. Heritability with low genetic advance in percent of mean was observed in seed width which indicated that non-additive gene effects were involved for the expression of this character and selection for such trait might not be rewarding. High heritability with high genetic advance in percent of mean was observed for pod width, inflorescence length indicated that this trait was under additive gene control and selection for genetic improvement for this trait would be effective. Correlation studies revealed that the highest significant association of yield per plant was observed with pod length, pod weight, pods per plant, inflorescence per plant. Path co-efficient analysis revealed the maximum direct contribution towards yield per plant was with pod weight followed by pods per plant, pod width and number of flower per inflorescence. The highest intra-cluster distance was found in cluster IV and lowest in cluster 1. Among five clusters, the highest inter-cluster distance was observed between cluster I and cluster TI and the lowest between cluster Ill and cluster IV. Considering all the characters the 67 (1313-8832). The objectives of the study were to measure the variability among the genotypes for yield and yield contributing characters, estimate genetic parameters, association among the characters and their contribution to yield. There was a great deal of significant variation for all the characters among the genotypes. High genotypic co-efficient of variation (CCV) was observed for yield per plant, number of male flower and number of fruits per plants whereas low genotypic co-efficient of variation (GCV) was observed for weight of fruit and breadth of fruit. In all eases, phenotypie variances were higher than the genotypic variance. High heritability with tow genetic advance in percent of mean was observed in days to first female flowering which indicated that non-additive gene effects were involved for the expression of this character and selection for such trait might not be rewarding. High heritability with high genetic advance in percent of mean was observed for yield per plant and number of fruits per plants indicated that this trait was under additive gene control and selection for genetic improvement for this trait would be effective. Goi, (BD-4256), (37 (CU937F1). 009 (13D-4309) were selected for future breeding programme.Item VARIABILITY STUDIES IN F4 PROGENIES OF Brassica rapa OBTAINED THROUGH INTERVARITAL CROSSES(DEPARTMENT OF GENETICS AND PLANT BREEDING, SHER-E-BANGLA AGRICULTURAL UNIVERSITY, DHAKA, BANGLADESH, 2010-06) ALAM, MD. FAYZULA research was conducted by using twenty six (26) F.1 populations of some inter-varietal crosses of Brassica rapa and grown in the experimental farm of Sher-e-Bangla Agricultural University. Dhaka, during November 2009 - March 2010 to study the magnitude of variations in characters, heritability, genetic advance, character associations, direct and indirect effect of different characters on seed yield. There were significant variations in number of primary branches per plant, number of secondary branches per plant, number of silliqua per plant, days to 50% flowering, length of siliqua, number of seeds per siliqua, 1000 seed weight and yield per plant showed least difference between genotypic and phenotypic variances. Plant height, length of siliqua, number of silliqua per plant, days to 50% flowering showed low genotypic and phenotypic coefficient of variation. Plant height, number of primary branches per plant, number of secondary branches per plant and number of silliqua per plant showed high heritability coupled with high genetic advance and very high genetic advance in percentage of mean. However, length of siliqua showed low heritability. Correlation study revealed that yield per plant had significant positive association with plant height. number of primary branches per plant, number of siliqua per plant, seeds per siliqua. and siliqua length (genotypic or phenotypic level). Path co-efficient analysis revealed that plant height, number of primary branches per plant, number of siliqua per plant, seeds per siliqua, and siliqua length had the positive direct effect on yield per plant and days to 50% flowering, number of secondary branches per plant, and thousand seed weight had the negative direct effect on yield per plant. Based on the variability study, some F4 plants showing high heritability for short duration and yield contributing characters were selected from some of cross combinations of the intervarital crosses of Brassica rapa for further selection.Item GENETIC DIVERSITY, CORRELATION AND PATH ANALYSIS IN RICE (Oryza saliva L.)(DEPARTMENT OF GENETICS AND PLANT BREEDING, SHER-E-BANGLA AGRICULTURAL UNIVERSITY, DHAKA, BANGLADESH, 2010-12) CHAKMA, SHYAMA PRASHADThe experiment was conducted at the experimental field of Shcr-e-Bangla Agricultural University. Dhaka under the agro-ceological zone of Modhupur l'ract (AEZ-28) during January to June of 2011. The experiment was intended to estimate the nature and magnitude of genetic variability, correlation, path coefficient and genetic diversity of 39 rice genotypes collected from Bangladesh Rice Research Institute, Lal Teer Seed Ltd. and Chittagong lull Tracts. All the characters tested showed significant variations among the genotypes. High genotypic coellicient of variation was observed for grain yield and number of unfilled grains per panicle. I ugh heritability coupled with high genetic advance in percent of mean was observed in grain yield, harvest index. 1000-grains weight, unfiLled grains per panicle. secondary branches number and plant height indicating additive gene control on the expression of these characters and that selection based on these characters might be effective. The significant positive correlations were recorded for number of effective tillers per plant. 1000-grains weight and harvest index with grain yield at both genotypic and phenotypie levels indicating that the improvement of these characters would lead to increase in grain yield. The highest positive direct effect on grain yield for harvest index followed by number of filled grains indicated that the direct selection based on these characters would be effective for yield improvement. The genotypes were grouped into six clusters. Cluster I consisted of the highest II genotypes from different origin, whereas cluster IV consisted of the lowest three genotypes. The genotypes of Chittagong llill Tracts were distributed in different clusters indicated that the geographical distribution was not related to genetic diversity. The highest inter genotypic distance was observed between Tharak c/han and BRRI dhan 45 and the lowest between BRRI dhan 33 and BRRI dhan 50. The highest inter cluster distance was observed between cluster II and Ill and the highest intra cluster distance for cluster IV. Cluster III had the highest mean values for grain yield, harvest index. 1000-grains weight and number of effective tillers and cluster V for number of primary branches and cluster VI for number of tilled grains that had significant positive correlation with grain yield. Considering all the data analyses, the genotypes BRRI dhan 36. Gold, BRRI dhan 28, Dowel, BRRI dhan 45, BRRI dhan 50 and indian Pajani could be selected for hybridization program.Item CALLUS INDUCTION AND IN VITRO PLANT REGENERATION OF BARI RELEASED POPULAR POTATO (Solanum tuberosuin L.) CULTIVARS(DEPARTMENT OF GENETICS AND PLANT BREEDING, SHER-E-BANGLA AGRICULTURAL UNIVERSITY, DHAKA, BANGLADESH, 2010-12) MOON, NUSHRAT JAHANThe present study was undertaken in the Genetics and Plant Breeding Laboratory and the 'lissue culture laboratory of the Department of Biotechnology. Sher-e-Baiigla Agricultural University, Sher-e-Bangla Nagar. Dhaka- 1207. during the period of January 2011 to December 2011. to investigate the in vitro perkrmance ot three Bangladesh Agricultural Research Institute ( BARI) released popular potato eultivars viz. Granola. Diamant and Asterix using potato sprout as expLant. live Levels of' (A (50. 100. 200. 300 and 400 ppti/l.) were used to find out the best concentration for sprout initiation, and it was revealed that (ib\ at the rate of 400 ppm/I. was the best concentration for maximum sprouting within short period of time. The effect of ditThrent combination and concentration of NAA and BAP on in vitro callus induction and plant regeneration was also studied. the concentrations for NAA were 1.0 mg/I. and 1.5 mg/!. and fOr BAP were 0.5 mg!L .. 1.0 mg/L. 1.5 mg/I. and 2.0 mg/L. The combined ctThet of both the honnones were also studied. the variety Diamant produced callus in the shortest period of time (4.46 days). It took minimum days 3.53 days) for callus induction in the treatment of' LU mulL NAA f 1.5 mg/I. BAP. The largest size (2.10 cm) and the highest weight (2.54 gm) of callus were noticed in the variety Diainant with the treatment 1.5 mg/L NAA within 28 DA I. In ease of shoot initiation, variety Granola took minimum time (13.17 days) followed by Diamant (1 3.63 days) and Asterix (15.01) days). The variety Granola produced the maximum number of' shoots'plantlet (3.00 shoots/plantlet) and the Longest (6.76 cm) shooise'plantlet ibllowed 1w Diamant (2.12 shoots/plantlet and 6.51 cm) and Asterix (1.91 shoots.'plantlet and 5.68 cm). respectively. Variety Granola also produced the maximum number of leaves/plantlet (20.0 leaves) with the treatment 1.5 mg/I.. NAA+l .5 mg/I. BAP within 28 DAI. The variet Asterix took the minimum (12.73 days) days to root initiation followed by Granola (15.01 days) and Diamant (15.48 days). In treatment 1.5 mg/I. NAA-0.5 mg/I. BAP required the minimum (0.93 das) days to root initiation and also the number of roots/plantlet (1 4.53 roots/planilet at 28 DAI ) was the maximum. When variety and treatment took tinder consideration, the treatment 1.5 mg/L NAA±0.5 mg/L. HAl' with variety Dianiani produced the maximum (19.40 roots/plantlet) and Granola produced the longest 14.42 cm) roots/plantlet respectively. The regenerated plantlets were transIrred successfully in plastic tray and subsequently to soil. The variety Graiola survived at high rate in growth chamber and in open atniosphere. Among all the varieties. Granola showed the best pertonnanee from callus induction, plant regeneration to in vivo establishment.Item MERISTEM CULTURE AND IN VITRO PLANT REGENERATION OF SOME POTATO (Solanum tuberosum L.) GENOTYPES(DEPARTMENT OF GENETICS AND PLANT BREEDING, SHER-E-BANGLA AGRICULTURAL UNIVERSITY, DHAKA, BANGLADESH, 2010-12) AHMED, SHAMMANAThe experiment was undertaken with a view to standardize a protocol for in v/tm culture and plant regeneration using potato meristem as explants. The experimental materials were six potato genotypes viz, Shil Bilati, Kufri Shinduri, Lal Pakri, Shil Pakri, Jam Alu and Pahari Pakri. Four levels of GA3 (100, 200, 300 and 400 ppm) were used to assess the influence of GA3 on sprouting abilities of six potato genotypes. The maximum sprouting efficiency was observed in 400ppm 0A3 treatment. Combinations of GA3, 2, 4-D and Kinetin were used along with fresh MS media to inoculate meristems and to induce callus. Maximum size of callus was observed (0.82 cm) in Kufri Shinduri inoculated in T4 (0.5 mg/I GA3 +1.0 mg/I 2, 4-0+ 1.5 mg/I KIN). Shil Pakri meristem inoculated in hormonal treatment T3 (0.5mg/i GA3 + 0.5mg/I 2, 4-D + 1.5mg/I KIN) showed best results regarding highest single shoot length/plantlet (5.88 cm). Three levels of GA3 treatments (fresh MS, MS+0.5mg/l GA3 and MS+1.5mg/1) were used to study subsequent shoot elongation and plant regeneration of single shoot/plantlet produced from meristem culture. Treatment T2 (0.5 mg/I GA3) gave maximum plant height (5.68 cm), number of branches/plantlet (2.60), root lengthfplantlet (8.74 cm) and number of leaves/plantlet (14.86). Shil Pakri gave maximum performance in respect of plant height (8.96 cm), number of branches/plantlet (2.78), number of roots/plantlet (4.33) and number of leaves/plantlet (22.56). Shil Pakri treated with T2 (0.5 mg/I GA3) showed maximum plant height (9.50 cm), number of branches/plantlet (5) and number of leaves/plantlet (33.67). Among all genotypes Shil Bilati was least responsive whereas Shil Pakri showed better overall performance from meristem culture to establishment of plants in field conditions. Plantlets obtained from meristem culture were tested using DAS-ELISA protocol for confirming plantlets to be virus free.
