Item CALLUS INDUCTION AND PLANT REGENERATION IN ONION (Allium cepa L.) FROM SHOOT TIP EXPLANT(Sher-e-Bangla Agricultural University Research System (SAURES), Dhaka, Bangladesh, 2009-01) Rashid, M. H.; Khalekuzzaman, M.; Hossain, M. S.; Rahman, M. S.; Hasan, M. F.An efficient protocol for callus induction and plant regeneration was established from shoot tip explant in onion (Allium cepa L.). Shoot tip from field-grown onion bulbs of Taherpuri and Indian cultivars were used as experimental materials in this investigation. Different concentrations of 2, 4-0 or NAA were used in MS medium for callus induction. The highest percent (86.6%) of callus induction was 86.6 in Taherpuri and in Indian (80%) cultivar in MS media containing 1.5 mg/I of 2,4-0 within fourteen to sixteen days of culture. Different concentrations and combinations of cytokinins (BAP and KIN) and auxins (NAA and lBAl were used for primary establishment of shoot tip. Maximum shoot proliferation was obtained after subculturing the callus in MS medium supplemented with 1.5 mg/l KIN + 1.5 mg/I of BAP within 20 days of callus culture. The highest percentage (100%) of root induction was achieved in Taherpuri cultivar on medium supplemented with 1.0 mg/l of NAA from in vitro raised shoots. Regenerated plantlets were successfully transferred on to the natural condition and showed healthy growth.Item CORRELA TION AND PATH CO-EFFICIENT ANALYSIS OF YIELD CONTRIBUTING TRAITS IN ADVANCED LINES OF Brassica rapa L.(Sher-e-Bangla Agricultural University Research System (SAURES), Dhaka, Bangladesh, 2014-07) Hussain, M. The significant positive correlations with seed yield per plant were found in thousand seed weight. no. of siliqua per plant. no. of primary branches per plant. Path co-efficient analysis revealed that plant height. no. of primary branches per plant, no. of siliqua per plant, siliqua length, thousand seed weight showed positive direct effect with yield per plant. Days to 50% flowering, days to 80% maturity. no. of secondary branches per plant, no. of seed per siliqua showed negative direct effect on yield per plant. S.To evaluate the effect of some chemicals and botanicals on jassid, Amrasca biguttula biguttula Ishida in brinjal and their impact on spiders and lady bird beetles. a field experiment was conducted at the experimental farm of Sher-e-Bangia Agricultural University. Dhaka during February to August 2007. The experiment comprised nine treatments viz. irnidachloprid 200 SL. cypermethrin 10Ee. carbosulfan 20EC, thiarnethoxarn 25WG. neern seed kCI11e1.neern oil combined with trix, trix, mahua leaf extract and an untreated control. It was laid out in a randomized completely block design with 3 replications. The lowest population of adult (2.36 plane' at vegetative stage and 9.80 plant" during fruiting stage) and nymph (0.73 plant" at vegetative stage and 1.67 plant" during fruiting stage) was observed in carbosulfan treated plots. which gave the maximum protection of brinjal against jassid attack by reducing pest population. Among the botanicals tested, neern oil in combination with trix, and neern seed kernel showed satisfactory control ofjassid on brinjal. Trix and mahua leaf extract reduced the infestation of jassid in comparison with control but their effectiveness was not satisfactory. Carbosulfan, imidachloprid and cypermethrin reduced more than 80% population of spiders and lady bird beetles.Item GENETIC DIVERGENCE IN OLEIFEROUS Brassica species(Sher-e-Bangla Agricultural University Research System, Sher-e-Bangla Agricultural University, Dhaka, Bangladesh, 2008-01) Zahan, M. I.; Bhuiyan, M. S. R.; Hossain, M. S.Genetic divergence among 41 genotypes of rapeseed and mustard was estimated using D' analysis. In each case of three species B. napus, B. rapa and B. juncea the genotypes under study fell into six clusters. The inter-cluster distances were larger than the intra-cluster distances in each species suggesting wider genetic diversity among the genotypes of each species. In Brassica napus Cluster II had the highest intra-cluster distance. Cluster I and III had no intra cluster distance because only one genotype was under each of the both clusters. Inter cluster distance was maximum between clusters II and IV. In Brassica rapa Cluster II had the highest intra-cluster distance and Clusters I, V andVl had the lowest intra cluster distances. Inter cluster distance was maximum between clusters I and IV. In Brassica juncea Cluster I had the highest intra-cluster distance while clusters II and VI had the lowest or no intra cluster distance. Inter cluster distance was maximum between clusters I and IV. The results revealed that genotypes chosen for hybridization from clusters with highest distances would give high heterotic F, and broad spectrum of variability in segregating generations.Item GENETIC VARIABILITY AND CHARACTER ASSOCIATION OF CHILLI (Capsicum frutescens L.)(Sher-e-Bangla Agricultural University Research System (SAURES), Dhaka, Bangladesh, 2010-07) Sultana, N.; Hossain, M. S.; Bhuiyan, M. S. R.Thirty chilli genotypes were studied with a view to find out variability and genetic association for fruit yield and its component characters. All the characters showed significant variation among the genotypes. The phenotypic variance was higher than the corresponding genotypic variance for all the characters. The highest genetic variabi I ity was obtained in number of fruits/plant followed by fruit yield, plant height and fruit length. High heritabil ity together with high genetic advance in percentage of mean was observed in fruit number/plant followed by fruit yield and plant height. Genotypic correlation coefficients were higher than the corresponding phenotypic correlation coetlicients for most of the traits. Fruit yield was positively and significantly associated with plant height, days to first flowering, number of primary branches/plant, fruit length. fruit weight and fruits number/plant.Item GENETIC VARIABILITY, CHARACTER ASSOCIATION AND PATH ANALYSIS OF WHITE JUTE (Corchorus capsularis L.)(Sher-e-Bangla Agricultural University Research System (SAURES), Dhaka, Bangladesh, 2010-01) Al-Mamun, M.; Hossain, M. S.; Khatun, R.; Yahiya, A. S. M.; Islam, M. M.ute (Corchorus capsularis L.) is an important fibre crop of manifold utility. Its fibre yield and quality is dependent on component characters which are quantitatively inherited and considerably influenced by environment. In crop plants, most economical characters are controlled by many genes. Physiological and linkage relationship have been frequently observed in genes governing various quantitative characters (Dhillon and Gupta, 1975). Hence, knowledge on the phenotypic and genotypic correlations between important characters is considered useful in planning an efficient breeding programme. Besides, correlations between important and unimportant characters may reveal that some of the latter are useful as indication of one or more of the important characters (Johnson et al., 1955 a). Thus, advancement through a breeding programme depends upon the extent of genetic variability in the population and the extent to which the desirable traits are heritable. In this context, correlation and path-coefficient analysis assume special importance as they not only measure mutual association but also estimate the direct and indirect contributions of perspective traits. This investigation was, therefore, undertaken to measure the genetic variability, nature of association among different plant traits and the potential applicability of path analysis techniques in breeding white jute.Item In Vitro PLANT REGENERATION FROM MESTA (Hibiscus sabdariffa L.)(Sher-e-Bangla Agricultural University Research System (SAURES), Dhaka, Bangladesh, 2008-07) Roy, T. K.; Khatun, A.; Hossain, M. S.; Mondal, R.An experiment was conducted in the Genetic Engineering Laboratory of the Cytogenetics Department. Bangladesh .lute Research Institute (B.lRI), Dhaka. Cotyledons (with attached petioles) of mesta (Hibiscus snbdnrifJa L.) were used as explant to investigate its in vitro regeneration potentiality. Seed germination percentage was found to be better on clinical cotton supported MS liquid medium (90%) compared to agarsupported MS solidified medium (81.33%). Among the combinations, MS medium supplemented by BAP (2.0 mgll) and IAA (0.5 mgl1) showed the highest (58.33%) shoot regeneration. Shoot regeneration was not observed in MS media without hormone (MSO) and also when more than 4mglL BAP with 0.5 rng/l IAA combination was used. MS medium without hormone (MSO) showed good response for root formation from regenerated shoots of mesta and root development occurred very nicely. The regenerated plantlets of mesta subsequently survived in soil.Item INTERGENOTYPIC VARIABILITY STUDY IN 58 ADVANCED LINES OF Brassica rapa(Sher-e-Bangla Agricultural University Research System (SAURES), Dhaka, Bangladesh, 2009-01) Mahmud, M. S.An experiment was carried out with 55 genotypes of Brassica rapa along with three commercially cultivated varieties as check to study their intergenotypic variability, correlation and path co-efficient analysis In respect of 10 different morphological characters at the experimental farm of Sher-c-Bangla Agricultural University. Dhaka during November 2007 to March 2008. Significant variation was observed among all the genotypes for all the characters studied except thousand seed weight. High GCY value was observed for number of secondary branches per plant. High heritability values along with high genetic advance in percentage of mean were obtained for days to 50% flowering, number of secondary branches per plant, seeds per siliqua and siliqua length. Highly significant positive association of seed yield per plant was observed with number of primary branches per plant, number of secondary branches per plant, number of siliquae per plant. Path analysis showed that yield per plant had the highest direct effect on number "I' primary branches per plant, number of siliquae per plant, number of secondary branches per plant and number of seeds per siliqua. Thus, these components were found as the important yield contributing traits and some lines were selected for future hybridization program to improve the yield status of the existing Brassica rapa genotypes.Item N VITRO PLANT REGENERATION FROM INTERNODE AND LEAF SEGMENTS DERIVED CALLUS IN Adhatoda vasica NEES.(Sher-e-Bangla Agricultural University Research System (SAURES), Dhaka, Bangladesh, 2009-07) Rahman, M. H.Ile investigation was carried out 10 observe callus induction and subsequent regeneration potentiality of Adhatoda vasica NEES from internode and leaf segments of mature plants. The nature of callus was hard in texture and greenish white in colour in all concentrations of 2,4·D. On the other hand, the nature of callus was loose in texture and creamy in colour in all concentrations of NAA which was achieved from internode and leaf segments on MS medium supplemented with 4.0 mg/l NAA within four weeks of inoculation. The callus produced large number of shoots when cultured on MS medium with 1.0 mg/l BAP +0.1 mg/l NAA within 4 weeks of subculture. In vitro raised shoots were rooted on MS medium supplemented with 1.0 mg/l IBA within 4 weeks of subculture. III vitro grown plantlets with well root system were successfully established in natural condition through successive phages of acclimatization. The regenerated plants were healthy, uniform and identical to the donor plants and the survival percentage was 80.Item Nutritional composition, fatty acids, bioactive compounds, and antioxidant activity of Nigella sativa seed grown in Bangladesh(Discover Food, 2024-07-04) Hossain, M. S.; Rahman, M. Z.; Mozumder, S. N.; Rahman, J.Black cumin seeds are extensively utilized for foods, cosmetics, and medicinal purposes. The current investigation was undertaken to analyzse the nutritional values, fatty acid compositions, bioactive compounds, and antioxidant roles of diferent black cumin genotypes available in Bangladesh. The genotype BSK-2074 had a higher content of proximate composition of crude oil (35.17%), crude protein (23.51%), total carbohydrate (35.23%), moisture (6.54%), ash (3.74%), crude iber (8.12%), and gross energy value (644.88 kcal/100 g) than the released varieties and cultivars of black cumin available in Bangladesh. The major fatty acid proile of the seed-oil of the BSK-2074 genotype revealed that the oil contained a high amount of linoleic (59.39%), oleic (25.72%), palmitic (13.11%), and stearic (2.25%) acids, as well as a low amount of arachidic, linolenic, myristic, behenic, palmitoleic, lignoceric, and eicosadienoic acids. Moreover, the genotype BSK-2074 contained a high amount of unsaturated (UFAs, 85%), and low content of saturated (SFAs, 18%) fatty acids, and the ratio of UFAs to SFAs was higher (4.39–5.37). Again, the seeds of the genotype BSK-2074 were also enriched in diferent minerals, viz., calcium, magnesium, potassium, phosphorus, iron, and zinc. Moreover, total phenolic contents (TPC) and lavonoid contents (TFC) were found signiicantly higher in the genotype BSK-2074 (478.47 ± 7.33 mg GAE/100 g and 284.07 ± 2.08 mg QE/100 g, respectively) than the local Kalozira variety (316.48 ± 2.33 mg GAE/100 g and 120.53 ± 3.57 mg QE/100 g) found in Bangladesh. The activities of antioxidants 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radicals and ferric reducing antioxidant power (FRAP) were found signiicantly higher in BSK-2074 (37.93% and 129.65 ± 0.19 mg GAE/100 g), than the local Kalozira variety (98.09% and 193.75 ± 0.19 mg GAE/100 g). Thus, the new genotype, BSK-2074, was identiied and suggested as the most promising and nutritionally enriched genotype of black cumin.Item VARIABILITY STUDY IN F2 PROGENIES OF THE INTER-VARIETAL CROSSES OF Brassica rapa(Sher-e-Bangla Agricultural University Research System (SAURES), Dhaka, Bangladesh, 2008-07) Parveen, S.; Bhuiyan, M. S. R.; Hossain, M. S.; Rashid, M. H.An experiment was carried out by using the F, population of 12 inter-varietal crosses. including reciprocals, of the species Brassica rapa for estimating the magnitude of variations in characters. heritability and genetic advance. There were significant variations among different F, materials used in the experiment. Genetic parameters viz. number of primary branches/plant, number of secondary branches/plant. length of siliqua, number of seeds/siliqua. days to 50% flowering, 1000 seed weight and yield/plant showed least difference between genotypic and phenotypic variance. Plant height, length of siliqua, number of seeds/siliqua and days to 50% tlowering showed low genotypic and phenotypic co-efficient of variation. Number of primary branches/plant and secondary branches/plant showed high heritability coupled with high genetic advance and very high genetic advance in percentage of mean. However, the yield/plant, days to maturity and length of siliqua showed low heritability. Based on the variability study, some promising F, plants showing high heritability for yield contributing characters were selected from some of the cross combinations of the inter-varietal crosses of Brassica rapa for advancing the generations.
