    CALLUS INDUCTION AND PLANT REGENERATION OF STEVIA (Stevia rehaudiana B.)

    The present research work was carried out in Biotechnology laboratory of the Department of Biotechnology. Sher-e-Bangla Agricultural University. Shcr-e-Bangla Nagar, Dhaka- 1207 from the period of January 2014 to December 2014 to evaluate the effects of different plant growth regulators in in vitro on shoot proliferation and root lbrmation and finally to develop a viable micropropagation protocol in Stevia (Stevia rehaudiana) using shoot tip and nodal segments as explants. Explants was inoculated in MS (Murashige and Skoog) media supplemented with the Benzvladenine (BA) and Kinetine (KIN) for direct shoot formation. Combination of BA and Indolebutyricacid (IBA) as well as combination of KIN and IBA were used in MS medium for indirect shoot formation via callus induction. The highest percent of shoot induction (83.42%) obtained on 2.0 mg/L BA and the minimum days to shoot induction (10.6) were also obtained on 2.0 mg/I.. BA. The highest 10.8 shoots and 17.2 leaves per explants also observed in BA 2.Omg/L. But maximum average length of shoots per explants (3.94 cm) were reported from media containing 1mg/I. KIN. Highest callus induction potentiality (83.58%) was observed in 2 mg/L BA+1.5 mg/L IBA The minimum days to callus induction (13.4) were recorded in 1.0 mg/L KIN+I.5 mg/L IBA but the minimum days to shoot induction (9.6) from callus were recorded in 2.0 mg/L BA+1.5 mg/L IBA. Righest percent of root induction (78.18%) with 7.2 roots per explants and minimum 12.6 days to root induction were noticed on 1.0 mg/I. IBA. The combined ellèct on percent of' root induction was highest (76.92%) in the treatment of 1.0 mg/L KIN t 0.5 mg/L IBA. The same treatment showed minimum days (13.4) to root induction and maximum number of roots per explants. In acclimatization 90%, 80% and 80% survival rate were recorded in growth cabinet, shade house and field condition respectively. In vitro regeneration technique was found to be very effective and promising niethod in the proliferation of Stevia and this method can be used 11w large scale production of disease free and quality planting material.

    CONTRIBUTION OF HOMESTEAD AGROFORESTRY PRACTICETOWARDS REDUCING POVERTY OF JALOKATHI DISTRICT

    Homestead Agroforestry is an important consideration for socioeconomic development in our country and also all over the world. Systematic Agroforestry practice is being popular day by day in Jalokahti district of Bangladesh. Considering the situation, the present study aims to contribution of homestead Agroforestry practice towards reducing poverty of Jalokathi District and explore the contribution of the selected characteristics of the homestead farmers on homestead Agroforestry practice. Data were collected by purposive random sampling method of 60 respondents from 350 farmers of three villages of Jalokathi union of Jalokathi Sadar upazila under Jalokathi district by using a pretested interview schedule during the period of 15 June 2017 to 15 December, 2017. Data were analyzed by SPSS version 16. Findings indicated that near about half (48%) of the respondents had high knowledge on homestead production and high Agroforestry contribution (household income) (52%) to reduce poverty under managed Agroforestry. Unmanaged Agroforestry also contributed to reduce poverty (77% with medium household income) but lower than managed Agroforestry system. Out of 8 selected characteristics of the respondents, educational qualification, homestead size, current housing condition, current household assets, current sanitation system and knowledge on Agroforestry had positive significant relationship with Contribution of homestead Agroforestry (i.e. household income) towards reducing poverty. The rest of the variables namely: age and family size did not show any significant relationships with Contribution of homestead Agroforestry (i.e. household income) towards reducing poverty.

    FINANCIAL PROFITABILITY AND RESOURCE USE EFFICIENCY OF JUTE CULTIVATION IN MADARIPUR DISTRICT OF BANGLADESH

    The present study was conducted to estimate financial profitability & resource use efficiency of Jute production in Madaripur district of Bangladesh. Two villages of Madaripur district under Madaripur sadar upazila were purposively selected for the present study. A total of 80 samples were collected for this study. The study is mainly based on primary data which were collected by the researcher herself through direct interviewing the sample farmers. Cobb-Douglas production function was used to satisfy the objectives. For calculating resource use efficiency, seven factors like cost on human labor, cost of seed, cost of urea, cost of TSP, cost of MP, cost of pesticides and cost of irrigation were considered. The jute producers had the opportunity to increase their productivity as well as profitability by proper utilization of these resources. In this study, Per hector variable cost and total cost of jute production were Tk. 50380 and Tk. 70380 respectively. The average gross return and net return were Tk. 128500 Tk. and 58120 per hactor respectively. Undiscounted BCR was 1.82 which implies that Tk. 1.82 would be earned by investing Tk. 1.00 for jute production. Gross margin was found 2.55. So, the Jute farming was found to be profitable for farmers. Returns to scale is 1.1 which means that the production functions exhibit increasing returns to scale. An increasing amount of investment in this production would further increase the returns from the farm. The study revealed that Human labor cost, irrigation and pesticide cost has significant impact on Jute production. Jute production delivers higher returns to farmers, as evidenced by the findings of this study. Farmers in the survey stated that improved yield and income encouraged them to continue producing jute. The study also identified some of the problems of Jute at different stages of Jute production. Institutional supports from Government and Non-government organizations for credit, input and technology may be helpful to increase jute production as well as income of jute farmers. Finally, on the basis of findings some recommendations were made for the development of jute sector in Bangladesh.

    GROWTH AND YIELD OF SOYBEAN AS AFFECTED BY IRRIGATION AND WEED CONTROL METHOD

    A held experiment was conducted at Sher-e-Bangla Agricultural University. Dhaka during December, 2013 to March,2014 with a view to fmd out the influence of irrigation and weed control method on the growth and yield of Soybean cv. BAR! Soybean 6.The experiment was carried out with four(4) irrigation treatments viz, no irrigation (control), one time(at 20 DAS,two times (at 20 and 40 DAS), three times (at 20. 40, and 60 DAS), and Ibur weed management treatments i.e., no weeding (control), one time hand weeding(at 20 DAS). two times hand weeding (at 20 and 40 DAS) and chemical control by Whip Super® (Fenoxaprop-p-ethyl)0j75g ha at 20 DAS. Results showed different types of weed werefound to infest experimental fields, among them Ec/zinocii/va co/onuin (72. 19%). Lindernia procumbens (28.28%) and Gynodon dacty/on (14.25%) had the highest relative density. It is also noticed that Lindernia procumbens created dominancy throughout the crop growth period even at the later stage of crop. 'three times irrigation (13) gave the highest (1.628 t had) seed yield on the other hand two times hand weeding (Wz) gave the highest (1.556 t ha") seed yield. Interaction effects showed the highest (1.917 t ha") seed yield from the combination of three times irrigation andtwo times hand weeding (13W2). This was also observed that herbicide Whip Super® showed better performance to control grass weeds but failed to control Lindernia procumbens. However, crop plants treated with herbicide became mature one week earlier than other treated crop plants. Considering weed control method of herbicide Whip Super®mav be most economic for cultivation of soybean.

A field experiment was conducted at Sher-e-Bangla Agricultural University, Dhaka during December, 2013 to March,2014 with a view to find out the influence of irrigation and weed control methods on the growth and yield of Soybean cv. BARI Soybean 6.The experiment was carried out with four(4) irrigation treatments viz, no irrigation (control), one time(at 20 DAS) ,two times (at 20 and 40 DAS), three times (at 20, 40, and 60 D AS), and four weed management treatments i.e., no weeding (control), one time hand weeding(at 20 DAS), two times hand weeding (at 20 and 40 DAS) and chemical control by Whip Super® (75g/L Fenoxaprop-p-ethyl)@75g ha-1 at 20 DAS. Results showed different types of weed werefound to infest experimental fields, among them Echinochloacolonum (72.19%), Linderniaprocumbens (28.28%)and Cynodondactylon (14.25%) had the highest relative density. It is also noticed that Linderniaprocumbens created dominancy throughout the field the later stage of crop. Three times irrigation gave the highest (1.63 t ha-1) seed yield on the other hand two times hand weeding gave the highest (1.56 t ha-1) seed yield. Interaction effects showed the highest (1.92 t ha-1) seed yield from the combination of three times irrigation andtwo times hand weeding. This was also observed that herbicide Whip Super® showed better performance to control grass weeds but failed to control Linderniaprocumbens. However, crop plants treated with herbicide became mature one week earlier than other treated crop plants. Considering weed control cost application of herbicide Whip Super®found to be most economic for cultivation of soybean.

    GROWTH AND YIELD RESPONSES OF RAPESEED-MUSTARD VARIETIES GROWN WITH DIFFERENT SOWING TIMES

    An experiment was conducted at Sher-e-Bangla Agricultural University farm, Dhaka to investigate the growth and yield responses of rapeseed-mustard varieties grown with different sowing times during November, 2017 to April, 2018. The experiment consisted of two factors of which were Factor A: Two mustard variety viz., V 1 = BARI Sarisha-11 = BARI Sarisha-17 1 = 2 November, S 2 2 = 9 November, S 3 = 16 November, S = 23 November and S = 30 November. The experiment was laid out in Randomize Block Design -1 5 -1 -1 -1 -1 -1 -1 -1 -1 nd day of November th November ( 5 -1 1 -1 and the lowest one -1 November. BARI Sarisha-11 along with sowing date S produced 572.73 % higher mustard grain than BARI Sarisha-17 along with sowing date 30 November.     PREVALENCE OF MASTITIS IN GOATS, ISOLATION OF ETIOLOGICAL BACTERIA AND IDENTIFICATION OF THEIR ANTIBIOTIC RESISTANCE PROPERTIES

    Mastitis is one of the multifactorial and expensive diseases of the dairy animals as it not only reduces the milk production but also leads to detrimental changes in the milk composition. The aim of this work was to determine the prevalence of clinical and subclinical caprine mastitis with their associated bacterial pathogens and antibiotic sensitivity patterns during the period from January to June, 2017. A total of 120 lactating goats in Dinajpur district were randomly selected for this study and 240 milk samples from each udder half were screened for evidence of clinical mastitis by clinical examination of udder and subclinical mastitis by CMT (California Mastitis Test). The overall prevalence of clinical and subclinical mastitis was found to be 11.67% and 38.75%, respectively. Bacteriological examination of mastitic milk samples revealed that Staphylococcus spp. (32.5%) was the major etiological agent of caprine mastitis followed by Escherichia coli (22.5%), Pseudomonas spp. (12.5%), Klebsiella spp. (12.5%), Bacillus spp. (5%). Statistical results of the study showed that parameters like age, parity, litter size and teat lesions had significant effect on caprine mastitis (p< 0.05) but lactation stage had insignificant effect on mastitis (p> 0.05). Antibiotic sensitivity test studies of mastitic pathogens from goat milk revealed sensitivity to Levofloxacin, Gentamicin, Ciprofloxacin and Chloramphenicol. Antibiotic resistance was seen against Penicillin, Amoxicillin, Azithromycin, Cloxacillin, Erythromycin, Vancomycin, Amikacin, Cefixime and Cephradine at various degree; the antibiotic resistance me be due to indiscriminate use of antibiotics in diseases control. It may be concluded that consumption of infected goat milk by children may host antibiotic resistance bacteria, the antibiotic resistance gene may transfer to the normal micro-flora that may lead super infection.

    YIELD VARIATION, SUGAR AND ANTIOXIDANT CONTENT OF POTATO AS INFLUENCED BY VERMICOMPOST AND FOLLOWED BY PERFORMANCE AT AMBIENT STORAGE CONDITION

    The experiment was conducted at Sher-e-Bangla Agricultural University, Dhaka1207, during the period from November, 2014 to April, 2015 to evaluate the effect of variety and vermicompost on the yield, sugar and antioxidant activity of potato and their performance under ambient storage condition. The experiment consists of two factors, i.e., factor A:- Potato varieties (V4): V 1 :BARI TPS-1, V :BARI Alu-28 (Lady Rosetta),V 3 : BARI Alu-25 (Asterix)and V 4 2 :BARI Alu-29 (Courage); factor B:-Vermicompost level (M-4):M 1 : 0 t/ha, M 2 : 2 t/ha, M 3 :4 t/ha and M : 6 t/ha and was laid out in a split-plot design with 3 replications. The present study revealed that vermicompost had significant effect on most of the yield and quality contributing parameters. Results demonstrated that yield and other parameters increased with increasing vermicompost level. Among the sixteen treatment combination, Asterix with vermicompost at the rate of 4 t/ha produced the maximum yield (36.01 t/ha) but Asterix with vermicompost at the rate of 6t/ha showed the highest specific gravity, dry matter, glucose, sucrose, ascorbic acid, antioxidant and polyphenol content. These two combinations also showed little bit higher concentration of glucose and sucrose compared to those of other combinations.In case of ambient storage condition; dry matter, specific gravity, starch, polyphenol and ascorbic acid decreased with increasing storing period while weight loss, total soluble solid (TSS), glucose, sucrose and antioxidant content increased with increasing storing period of up to 60 days. Lady Rosetta and Asterix may store under ambient storage condition up to 60 DAS without imparting any significant quality losses just prior to sprouting of tuber. It may be concluded that, the potato growers of Bangladesh may apply vermicopmpost on their field at the rate of 6 t/ha for maintaining better yield and good processing quality. 4
