    Development of an Advanced DNA Biosensor for Pathogenic Vibrio Cholerae Detection in Real Sample

    Due to the epidemics of emerging microbial diseases worldwide, the accurate and rapid quantification of pathogenic bacteria is extremely critical. In this work, a highly sensitive DNA-based electrochemical biosensor has been developed to detect Vibrio cholerae using gold nanocube and 3-aminopropyltriethoxysilane (APTES) modified glassy carbon electrode (GCE) with DNA carrier matrix. Electrochemical impedance spectroscopy (EIS), cyclic voltammetry (CV), Fourier transform infrared spectroscopy (FTIR), scanning electron microscope (SEM) experiments were performed to interrogate the proposed sensor at each stage of preparation. The biosensor has demonstrated high sensitivity with a wide linear response range to target DNA from 10−8 to 10−14 (R2 = 0.992) and 10−14 to 10−27 molL−1 (R2 = 0.993) with a limit of detection (LOD) value of 7.41 × 10−30 molL−1 (S/N = 5). The biosensor also exhibits a selective detection behavior in bacterial cultures that belong to the same and distant genera. Moreover, the proposed sensor can be used for six consecutive DNA assays with a repeatability relative standard deviations (RSD) value of 5% (n = 5). Besides, the DNA biosensor shows excellent recovery for detecting V. cholerae in poultry feces, indicating that the designed biosensor could become a powerful tool for pathogenic microorganisms screening in clinical diagnostics, food safety, and environmental monitoring.

    DISTRIBUTION OF WIDTEFLY, Bemisia tabaci Gennadius ON MUNGBEAN IN MAJOR MUNGBEAN GROWING REGIONS OF BANGLADESHI

    The study was conducted in the three mungbean growing locations/regions of Bangladesh viz., Bansal. Jessore and Gazipur during March 2005 to March 2006 to explore the distribution of whitefly. Bamisin (abaci Genn. and the incidence of Mungbean yel/ow mosaic virus (MYMV) disease infection in mungbean (Vigna radiata) as well to study the climatic factor(s) as the basis for regulating them. The incidence of whitefly and MYMV infection were the maximum in Barisal (253.7 adults and 82.90 nymphs/20 plants, and 37.33% leaves and 57.20% plants infection, respectively) and the lowest in Jessore (39.0 adults and 10. I 9 nymphs120 plants, and 18.20% leaves & 16.50% plants infection, respectively). The rate of MYMV infection in leaves and plants were positively correlated with the incidence of both whitefly adult and nymph. The climatic factors were attributed to the variations of whitefly incidence as well as performance of mungbean in locations. Among three locations, macroclimatic temperature (3 I .5°C), RH (84.44%) and rainfall (1.47 mm) as well as microclimatic temperature (25.6°C) and RH (77.8%) within crop canopy of mungbean were the highest in Jessore, which were detrimental to the incidence of whitefly. Conversely, macroclimatic temperature (28.4°q and RH (76.44%); microclirnatic temperature (21.53°(') and RH (73.73%) were the lowest in Barisal, which played as the favorable factors lor whitefly population. Both macro and microclirnatic temperature. RH and rainfall were negatively correlated to the incidence of whitefly. Accordingly, the incidence of whitefly and MYMV infection were the maximum in Barisal and minimum in Jessore and intermediate in Gazipur. Thus yield attributes were better and higher yield (1160.6 kg/ha) was obtained in Jessore, which was followed by Barisal (866.7 kg/ha), while the yield was minimum (414.7 kg/ha) in Gazipur.

    ECO-FRIENDL Y MANAGEMENT OF SHOOT AND FRUIT BORER (Earias vittella FAB.) USING BOTANICALS AND BIO-CONTROL AGENT

    A study was conducted at the Sher-e-Bangla Agricultural University, Bangladesh during May 2011 10 August 20 II to evaluate the effectiveness of botanicals and parasitoid for the management of okra shoot and fruit borer (Earias vittella Fab.). The experiment was laid out in a Randomized Complete Block Design (RCBD) comprising eight treatments viz., TI ; Neem oil @ 4mlll of water sprayed at 7 days interval, T2; Neem seed kernel extract @50 g/l of water sprayed at 7 days interval, TJ; Neem leaf extract @ 200gll of water at 7 days interval, T. ; Trichogramma evanescens @ 0.5g1plot at 7 days interval, T, ; Trichogramma evanescens @ 0.25g1plot at + neem oil @ 4mlll of water sprayed at 7 days interval, T6 ; Trichogramma evanescens @ 0.25g1plot + neem seed kernel extract @ 50gll sprayed at 7 days interval, T7; Trichogramma evanescells @ 0.25g1plot + neem leaf extract @ 200gll of water at 7 days interval, Ts ; Untreated control. Among the treatments T, (Spraying of neem oil @ 4mlll of water + release of Trichogramma evanescens egg parasitoid @ 0.25g1plot at 7 days interval) performed the best in terms of percent reduction of shoot infestation (96.50), flower bud infestation (83.33%) and fruit infestation by weight (86.90%).

    N VITRO PLANT REGENERATION FROM INTERNODE AND LEAF SEGMENTS DERIVED CALLUS IN Adhatoda vasica NEES.

    Ile investigation was carried out 10 observe callus induction and subsequent regeneration potentiality of Adhatoda vasica NEES from internode and leaf segments of mature plants. The nature of callus was hard in texture and greenish white in colour in all concentrations of 2,4·D. On the other hand, the nature of callus was loose in texture and creamy in colour in all concentrations of NAA which was achieved from internode and leaf segments on MS medium supplemented with 4.0 mg/l NAA within four weeks of inoculation. The callus produced large number of shoots when cultured on MS medium with 1.0 mg/l BAP +0.1 mg/l NAA within 4 weeks of subculture. In vitro raised shoots were rooted on MS medium supplemented with 1.0 mg/l IBA within 4 weeks of subculture. III vitro grown plantlets with well root system were successfully established in natural condition through successive phages of acclimatization. The regenerated plants were healthy, uniform and identical to the donor plants and the survival percentage was 80.

    Sensitive MWCNTP-Cys@MIP Sensor for Selective Electrochemical Detection of Ceftizoxime

    An electrochemical sensor based on molecular imprinted polymer (MIP) to detect ceftizoxime (CFX) with high sensitivity and selectivity is demonstrated. MIP was synthesized by electropolymerization of poly-cysteine (P-Cys) on a multi-walled carbon nanotube (MWCNT) modified glassy carbon electrode (GCE). A targeted drug was used as a template molecule during the polymerization process. The bare GCE was coated with a layer of MWCNT before the synthesis of MIP to improve the sensor sensitivity. Experimental parameters such as polymerization conditions, the influence of pH, molar ratio of the template molecules and the monomer molecules were all optimized. The peak potential exhibited linearity with CFX concentration in the ranges of 1 ×10−9×10−9–1× 10−710−7 molL−1 (R2 = 0.9904) and 2×10−710−7–1× 10−410−4 molL−1 (R2 = 0.9949). The LOD of the MIP sensor was found to be 1×10−101×10−10 molL−1 under optimal conditions using a differential pulse voltammetry (DPV). The proposed sensor was tested on real samples, and good recovery results were obtained.

    STATUS OF FRUIT SEED TRADE IN BANGLADESH AND TRADERS' PRACTICE FOR SEED QUALITY

    Seed is the vital component in the production of quality planting materials (QPM). A survey was carried out in 260 upzilla under 52 districts of Bangladesh during 2005-06 to study the status of seed trade of different fruit species in the country. The survey revealed that more than 20 fruit species traded in 531 seed stores and a total of 16288.10 kg seeds were sold out amounting Tk. 35.90 million. Variations in different fruit seeds traded in surveyed districts of Bangladesh were also observed. Significant number of seed traders expressed their awareness about the deterioration of seed due to attack of pathogens but did not take any measurement to protect their seeds. The present investigation showed that considerable amount of seeds of fruit species are being traded every year. as such, the issue of fruit species demands attention of the concerned authorities to take necessary action.
