Abstract

The study on humoral immune response of IBD live vaccine on broiler chicken through the different route of vaccination was conducted on 100 birds which were purchased from local market. The birds were divided into 3 groups and each group consists of 30 birds. Birds of each group were vaccinated with IBD live vaccine through different routes (oral, ocular, nasal) to find out the suitable route by increasing immune response. Rest 10 birds were kept to monitor the persistence and bio-degradation of maternally derived antibody. These birds were not vaccinated with IBD live vaccine. Primarily birds were vaccinated at day 10 and boostered at day 17. Blood samples were collected on 10 days. All the samples were tested with commercially available ELISA kit. In the present study the IBD antibody ELISA kit was used to monitor the antibody induced by vaccination and maternally derived antibody. In case of ocular route it is observed that the titer before primary vaccination was (1373.89 to 1680.67) (1517.91 ± 62.96) this is the maternally derived antibody but at 17 th th th , 17 , 24 day after primary vaccination it increases significantly (p<0.01) which ranges (1812.64-2245.32) (2025.02± 69.90). The birds were boostered at day 17. After booster dose that is at the age of 24 th day the ELISA titer again increases (3385.78-3680.52) (3520± 50.07). In nasal route, 17 th days after primary vaccination it increases (1705.33-1813.32) (1771.24± 19.45) significantly (p<0.05). Again at the age of 24 th day it increase (2204.99-2724.76) (2533.44±88.54). But in oral route at 17 th day after primary vaccination it increases (2012.39-2547.48) (2211.35 ±92.26) significantly (p<0.05). Booster dose was administered at day 17 th , after booster dose that is at the age of 24 day the titer again increased (3080.44-4743.13) (3821.92± 314.8). Among these three routes, it was found that immune response was better but higher in oral route. From the present study it is concluded that the oral route of vaccination with live vaccine against IBD ensures maximum antibody production in comparison to ocular and nasal route of vaccination.
