    Anti-inflammatory and analgesic activities of acetophenone semicarbazone and benzophenone semicarbazone

    To study anti-inflammatory and analgesic activities in swiss albino mice, two schiff bases namely acetophenone semicarbazone (ASC) and benzophenone semicarbazone (BSC) were synthesized and characterized. Methods: Two doses of the test compounds 25 and 50 mg/kg (p.o) for each were selected throughout the research work. The anti-inflammatory activity of the test compounds was determined by 'carragenan induced mice paw edema inhibition' method. The analgesic activity was determined by both, 'acetic acid induced writhing' and 'tail immersion' methods. All such data were compared with standard drugs at the dose of 10 mg/kg (p.o.). Results: Both ASC and BSC have showed positive effects as anti-inflammatory and analgesic agents. Anti-inflammatory and analgesic activities of the test compounds at 50 mg/kg (p.o.) were quite comparable to those of standard drugs at 10 mg/kg (p.o.). Conclusion: Both ASC and BSC can be considered as potent anti-inflammatory and analgesic agents.

    Anticancer activity of alangium salvifolium flower in ehrlich ascites carcinoma bearing mice

    The research study was conducted to determine the antitumor effect of the flower of Alangium salvifolium (crude extract and diethylether fractions) against Ehrlich Ascites Carcinoma (EAC) in mice at the doses of 10 mg kg-1 body weight intraperitoneally. Extract/fractions was administered for nine consecutive days. Twenty-four hours of last dose and 18 h of fasting, the mice were sacrificed and antitumor effect was assessed by evaluating tumor volume, viable and nonviable tumor cell count, tumor weight and hematological parameters of EAC bearing host. Significant (p<0.001) increases of survival times 30±0.96 and 25±0.40 days for crude extract and diethylether fraction of the A. salvifolium (10 mg kg-1) treated tumor bearing mice, respectively were confirmed with respect to the control group (20±0.13 days). The extract/fraction also decreased the body weight of the EAC tumor bearing mice. Hematological studies reveal that the heamoglobin (Hb) content was decreased in EAC treated mice whereas restoration to near normal levels was observed in extract treated animals. There was a significant (p<0.001) decrease in RBC count and increase in WBC counts in extract/fraction treated animals when compared to EAC treated animals. From the result it was showed that the extract has significant anticancer activity and that is comparable to that of Bleomycin.

    Antidiarrheal, antioxidant and antimicrobial activities of the musa sapientum seed

    Musa sapientum (M.sapientum) commonly known as 'banana' is widely used in Bangladeshi folk medicine for the treatment of various ailments including diarrhea. Hence, the present study was designed to investigate antidiarrheal, antioxidant and antibacterial potential of the methanolic extract of M.sapientum seed (MMSS). The extract was studied for antidiarrheal property using castor oil and magnesium sulfate induced diarrheal model and charcoal induced gastrointestinal motility test in mice. Total phenolic and flavonoids content, total antioxidant activity, scavenging of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, as well as nitric oxide (NO) and assessment of reducing power were used to evaluate antioxidant potential of MMSS. In addition, disc diffusion methods were used for antibacterial assay using various diarrheal induced bacterial strains. At the doses of 100 and 200 mg/kg body weight, the extract reduced the frequency and severity of diarrhea in test animals throughout the study period. At the same doses, the extracts significantly (p<0.001) delayed the intestinal transit of charcoal meal in test animals as compared to the control. In DPPH and NO scavenging method, MMSS showed good antioxidant potentiality in a dose dependent manner with the IC50 value of 12.32±0.33 μg/ml and 18.96±1.01 μg/ml, respectively with a significant (p<0.001) good reducing power. The extract also displayed strong antibacterial effect against when tested against Escherichia coli, Shigella dysenteriae, and Pseudomonas aeruginosa. Altogether, these results suggest that the MMSS could be used as a potential antidiarrheal agent along with its antioxidant and antibacterial potentiality.

    Antioxidant and antitumor activities of Polyalthia simiarum (Buch.-Ham. ex Hook. F. & Thomson) Hook. F. & Thomson

    The present study has been designed to investigate the antioxidant and antitumor activities of ethyl acetate (EA) extract of stem bark of Polyalthia simiarum (Annonaceae). Antioxidant potential of the EA extract was evaluated in vitro by DPPH (1,l-diphenyl-2-picrylhydrazyl) scavenging assay method. The EA extract showed free radical scavenging activity with IC50 value of 24.08 ± 0.30 fig/mL. The antitumor activity of the extract was determined against Ehrlich ascites carcinoma (EAC) in mice at 25 mg/kg and 5Í) mg/kg body weight intraperitoneal. Significant (p<0.001) increase of survival time by 25 ± 0.57 and 27 ± 0.40 days by the EA extract treated tumor bearing mice was confirmed with respect to the control group (22 ± 0.12 days). Hematological studies revealed that the hemoglobin (Hb) content was decreased in EAC treated mice whereas restoration to close to normal levels was observed in extract treated animals. There was a significant (p<0.001) decrease in RBC and increase in WBC counts in extract treated animals when compared to EAC affected animals. Therefore, the EA extract of P. simiarum was capable to exhibit moderate antioxidant and antitumor activities. This is the first report of antioxidant and antitumor potential of P. simiarum.

Materials and Methods: The antioxidant activity of methanolic extract of S. nodiflora (SN) was determined using Folin Ciocalteu reagent, phosphomolybdenum method, free radical scavenging activity by 1,1-diphenyl-2-picryl-hydrazyl (DPPH) and the reducing power activity. Analgesic activity was done by acetic acid and formalin model and CNS depressant activity was measured by the hole cross and open field method. Results: The total phenols and total antioxidant capacity of SN was found to be 37.38 < 2.01 mg/g equivalent of gallic acid and 433.37 < 4.22 mg/gm equivalent of ascorbic acid. The percentage (%) scavenging of DPPH free radical was found to be concentration dependent and IC50 value was 125.89 < 2.23 µg/ml while the IC50 value of standard ascorbic acid was found to be 16.76 < 0.11 µg/ml. The reducing power of SN was found to be concentration dependent. Acetic acid and formalin models are peripherally acting analgesic methods. The oral administration of both doses (100 and 200 mg/kg b.wt.) of SN significantly (p<0.001) inhibited 40.06% and 61.83% writhing response induced by acetic acid whereas oral administration of the same doses of SN significantly (p<0.001) inhibited 56.96% and 62.60% itching response induced by formalin. The methanolic extract of SN, at the dose of 250 mg/kg and 500 mg/kg b.wt. produced significant (P<0.001) decrease of locomotion. This is the first report of CNS depressant activity of the plant.

    Comparative study of antidiabetic activity of Cajanus cajan and Tamarindus indica in alloxan-induced diabetic mice with a reference to in vitro antioxidant activity

    Oxidative stress not only develops complications in diabetic (type 1 and type 2) but also contributes to beta cell destruction in type 2 diabetes in insulin resistance hyperglycemia. Glucose control plays an important role in the pro-oxidant/antioxidant balance. Some antidiabetic agents may by themselves have antioxidant properties independently of their role on glucose control. Objective: The present investigation draws a comparison of the protective antioxidant activity, total phenol content and the antihyperglycemic activity of the methanolic extract of Cajanus cajan root (MCC) and Tamarindus indica seeds (MTI). Materials and Methods: Antidiabetic potentials of the plant extracts were evaluated in alloxan-induced diabetic Swiss albino mice. The plant extracts at the doses of 200 and 400 mg/kg body weight was orally administered for glucose tolerance test during 1-hour study and hypoglycemic effect during 5-day study period in comparison with reference drug Metformin HCl (50 mg/kg). In vitro antioxidant potential of MCC and MTI was investigated by using 1, 1- diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity at 517 nm. Total phenolic content, total antioxidant capacity and reducing power activity was also assayed. Results: There was a significant decrease in fasting serum glucose level (P < 0.001), reduction in blood glucose level (P < 0.001) in 5-days study, observed in the alloxan-induced diabetic mice. The reduction efficacy of blood glucose level of both the extracts is proportional to their dose but MCC is more potent than MTI. Antioxidant study and quantification of phenolic compound of both the extracts revealed that they have high antioxidant capacity. Conclusion: These studies showed that MCC and MTI have both hypoglycemic and antioxidant potential but MCC is more potent than MTI. The present study suggests that both MCC and MTI could be used in managing oxidative stress.
