Abdus; Khalequzzaman, M.Visceral leishmaniasis (VL) or kala-azar is a neglected chronic systemic illness caused by Leishmania donovani. Clinical diagnosis of VL is inaccurate even in patients meeting the clinical case definition, so laboratory confirmation is essential. Current diagnostic tests include parasitological, immunological and PCR-based molecular methods. Except serology all other methods are not suitable as point-of-care test in remote endemic areas. Serological test by rK39 antigen is being used in the Indian subcontinent with very high sensitivity but it lacks discriminatory power between active VL and past or asymptomatic cases. So, always there is search for new diagnostic innovation which will be highly accurate, cheap, simple and rapid for successful VL management and to achieve the goal of current elimination programme. The present study was designed to formulate an applicable diagnostic algorithm based on comparative evaluation of available VL diagnostic tests aiming to facilitate more case detection from different levels of health care facilities in our country. The work also focused on exploration of genotype and genetic diversity of Leishmania strain for its taxonomical status. Study population comprised of 200 subjects including 100 confirmed VL patients (spleen smear positive for LD body) admitted in the Rajshahi Medical College Hospital (RMCH), Bangladesh and 100 controls (30 endemic, 30 non-endemic and 40 disease controls) from July, 2010 to January, 2013. Parasitological (spleen and buffy coat smear microscopy), serological (RDT and ELISA against both rK39 & rK28) and molecular diagnostic tests like Loop-mediated isothermal amplification (LAMP) and different PCR (Ln-PCR, Mini-exon, ITS1 and ITS2) methods were carried. Buffy coat was separated by concentration gradient separation technique to be utilized for smear preparation and DNA extraction. Parasite load in spleen smear was graded from 1+ to 6+ and performances of tools other than serology were compared with parasitic load. Diagnostic sensitivity, specificity and accuracy were calculated using online clinical calculator. Performances of different diagnostic tools against gold standard (spleen smear) were compared by McNemar’s test and correlations between tools were done by Spearman correlation. The kappa value of different tools against spleen smear was used to construct a receiver-operator characteristic (ROC) curve. Diagnostic tools were ranked (1 to 10) based on kappa value, cost, interpretation, availability, user friendliness, test type and potential for field use. Diagnostic algorithm for VL in Bangladesh was formulated based on the diagnostic indices and index scoring of tools. Genotyping by ITS1-RFLP and DNA sequence analysis of ITS1 and ITS2 PCR products was done for polymorphism. Phylogenetic tree was constructed using maximum parsimony by the MEGA (Molecular Evolutionary Genetics Analysis) computer program, Version 5.05 and compared with reference strains. Parasitic load was demonstrated as grade 1+ (8%), 2+ (41%), 3+ (34%), 4+ (10%), 5+ (6%) and 6+ (1%). Diagnostic sensitivity, specificity and accuracy at 95% CI respectively of rK39 ICT (99%, 96%, 95%), rK39 ELISA (98%, 97%, 95%), rK28 ICT (99%, 90%, 89%), rK28 ELISA (98%, 94%, 92%), Buffy coat smear (93%, 100%, 93%), Ln-PCR (94%, 99%, 93%), LAMP (89%, 100%, 89%), Mini-exon PCR (86%, 100%, 86%), ITS1 PCR (85%, 100%, 85%) and ITS2 PCR (80%, 100%, 80%) were noted. All diagnostic tools correlated significantly (P=0.0001) with gold standard. Correlation between rK39 and rK28 ICT was 0.942 (P=0.0001) and between rK39 and rK28 ELISA was 0.930 (P=0.0001). Pair-wise comparison of Buffy coat smear, Ln-PCR, LAMP, Mini-exon, ITS1 and ITS2 PCR against gold standard was found significant (P<0.05). Different PCR methods were also found significantly (P<0.05) comparable to Ln-PCR. Buffy coat smears was found 100% positive among grades ≥3+ parasite load cases but other tools did not show significant correlation with parasite load. ROC analysis in respect to gold standard revealed AUC (at 95% CI) of 0.965 (SE=0.0129), 0.945 (SE=0.0157), 0.930 (SE=0.0174), 0.925 (SE=0.0179) and 0.900 (SE=0.0201) respectively for Ln-PCR, LAMP, Mini-exon, ITS1 and ITS2 PCR. While, AUC (at 95% CI) of rK39 ICT, rK39 ELISA, rK28 ICT, rK28 ELISA and buffy coat smear was 0.975 (SE=0.0110), 0.975 (SE=0.0111), 0.945 (SE=0.0159), 0.960 (SE=0.0139), and 0.965 (SE=0.0128) respectively. Index score (IS) of diagnostic tools showed rK39 ICT as the best diagnostic option (Rank=1, IS=1.65335) followed by rK28 ICT (Rank=2, IS=1.48776), Buffy coat smear (Rank=3, IS=0.50764), rK39 ELISA (Rank=4, IS=0.2963), rK28 ELISA (Rank=5, IS=0.2963), Ln-PCR (Rank=6, IS= -0.7158), LAMP (Rank=7, IS= -0.88139), Mini-exon PCR (Rank=8, IS= -0.88139) ITS1 PCR (Rank=9, IS= -0.88139), and ITS2 PCR (Rank=10, IS= -0.88139) for kala-azar. In the diagnostic algorithm, RDT (rK39 or rK28) was proposed as the most preferred test. Genotyping revealed L. donovani as the sole agent for kala-azar and DNA sequencing explored no polymorphism. All 14 Bangladeshi strains including 4 from present study (Raj 21, Raj 64, Raj 68 and Raj 85) were found exactly identical to strain ID, DON 39, with Zymodeme MON-2, isolated from Indian VL patient (gene accession No. AJ634376). Kala-azar patients’ characteristics showed male to female ratio of 2.13:1 with mean age, monthly income and duration of fever as 20.66±15.863 years, Tk. 4005±2631.871 and 20.61±12.518 weeks respectively. Patients had splenomegaly (98%), hepatomegaly (76%), anaemia (99%), wasting (100%), blackening (96%), weight loss (100%), family history of VL (18%), past history of VL (13%), jaundice (2%), bleeding (9%) and pancytopenia (40%) as presenting features. All diagnostic tests showed good to excellent sensitivity (80% to 99%) and specificity (90% to 100%). rK39 ICT ranked #1 and was proposed as the most preferred diagnostic test in the diagnostic algorithm. Buffy coat smear may be suitable alternative to conventional invasive procedures and is preferred parasitological confirmation for asymptomatic, treatment failure, relapse or immunodeficient VL patients. Ln-PCR or LAMP can be good adjunct for confirmation of kala-azar. Genotyping revealed L. donovani as the agent of kala-azar in Bangladesh, identical to Indian strain (DON 39, Zymodeme MON-2) and there is no strain polymorphism.Item Effect of Mutagens and Antibiotics on Some Traits of The Ladybird Beetle Epilachna vigintioctopunctata (Coleoptera: Coccinellidae)(University of Rajshahi, 2015) Hoque, Kazi Md. Abdus; Zaman, ShahriarEffects of methyl methanesulfonate (MMS), ethyl methanesulfonate (EMS) and actinomycin-D (ACM-D) on the quantitative traits such as, dominant lethal mutation (DLM), number of eggs, number of larvae, number of pupae, sex-ratio, and incubation period, larval and pupal period and longevity of adult males and female were studied in Epilachna vigintioctopuntata. Four different doses of MMS, EMS viz. 1.0ml, 2.0ml, 3.0ml and 4.0ml were used, five types of crosses were designed and the experiments were carried out for three generations. In crosses of Epilachna vigintioctopuntata it was found that all the doses of MMS and EMS induced dominant lethal mutations. However, this effect was not significant in all the crosses. There was a linear relationship between the doses of mutagen and dominant lethal frequency. The mutagenic effect of actinomycin-D on the dominant lethal mutations revealed that it induced dominant lethal mutations (significant or not) but it produced weak effect in the induction. The effects of actinomycin-D on the frequency of MMS, EMS-induced dominant lethal mutations showed that actinomycin-D did not play any role to modify the effects of alkylating agent on the induction of dominant lethal mutations. Effects of MMS, EMS and actinomycin-D on the number of eggs, larvae and pupae were studied. It was observed that the number of eggs, larvae, and pupae were decreased in the treated crosses in comparison to the controls of the parental, F1 and F2 generations. It was also noted that the combined action of MMS or EMS and actinomycin-D significantly decreased the number of eggs, larvae and pupae in the parental generation. Mutagenic effects of MMS, EMS and actinomycin-D on the number of male and female were also studied. In the F1 and F2 generation numbers of male and female were decreased significantly in some treated crosses than in the controls. The incubation period was decreased in the treated crosses compared to the controls but the value was increased in only one treated cross in the parental generation. In the F1 and F2 generations, the incubation period was found to increase in some crosses than the controls. For the larval period, pupal period, longevity of male and female it was observed that in the parental, F1 and F2 generations, the larval period was increased for the treated crosses in comparison to the control cross. But larval period did not change in the cross ACM-D♂ x control♀. For the effects of MMS and EMS on the pupal period was found to gradually increase with doses. The longevity of adult males and females were also gradually increased by the increased mutagenic doses and antibiotic in three generations. I did my internship at one of the well-established organizations namely; DTech Online Limited as a Trainee under the tenure of “Network Support Engineer.” This is the reason I want to point out my experience and the knowledge I have gained during the internship. Shifting to my role, as a Network Engineer intern, I specifically aimed at mastering on networking, and gaining information about Switching, Routing, IP addressing, fiber connection solutions etc.; different types of routing and their relative protocols. Today, most of the network engineers are engaged with new technologies. This organization is provided with several gadgets that are linked with others through the interfaces. In the server room we have different Mikrotik routers, switches, OLTs, hubs, computers, printers and other important items. The ISP segment is to connect people and enterprises with the Internet and provide Internet services. I learned about the wiring of routers, Router Operating System for Mikrotik, switch configuration, routing, static and bridge configurations, installation of wireless routers, DHCP configuration, NAT configuration, bandwidth management, PPoE settings, IP addressing, fiber optic cable, local UTP cabling, and home and corporate router as well as switch installation. My conclusion of the internship is presented and highlights of the report are discussed in this section. This is why I opted to make this specific topic of my report: This internship made me to learn the practical aspect of working within the networking environments, and improve on my communication skills. In addition, an attempt was made to determine the degree of success of my internship.Item Intrinsic Features of Nonlinear Waves in Dusty Plasmas(University of Rajshahi, 2024) Salam, Md. AliDusty plasmas are electrically conducting ionized gases (macroscopically quasi-neutral) that comprise positively and negatively charged dust particles in addition to ions and electrons. Dusty plasmas exist in the Earth's magnetosphere, cometary tails, planetary rings, asteroid rings, rotating stars, and many other astronomical environments. In dusty plasmas, various types of nonlinear waves, such as solitary waves, shock waves, etc., may be propagated. The presence of dust particles makes the plasma system more complex. Besides, the characteristics of nonlinear waves can be substantially affected by different forces and plasma parameters. We theoretically investigate the characteristics of dust-ion-acoustic solitary and shock waves, where the plasma species follow different particle distributions. The higher-order nonlinear and dispersive (or dissipative) effects on the waves are examined. We also investigate various kinds of effects, such as magnetic, adiabatic, parametric, etc., while it is illustrated that how they change the wave characteristics. An inhomogeneous KdV-type or modified KdV-type equation is obtained for demonstrating the solitary waves, whereas an inhomogeneous modified Burgers-type equation is obtained for the shock waves. The reductive perturbation method is extensively used for incorporating the higher-order effects into the KdV, modified KdV, and modified Burgers equations. The re-normalization technique, the Abel’s theorem, and the method of variation of parameters are used for adding higher-order nonlinear and dispersive (or dissipative) effects into the solutions. We deal with the theoretical investigation of the combined effect of the magnetic field and plasma rotation on the nonlinear features of obliquely propagated dust-ion-acoustic solitary waves in a magnetized dusty plasma. From the investigation, it is found that the overall impact of the magnetic field, oblique rotation, electron temperature, and dust concentration has a crucial role in changing the amplitude, width, and phase speed of the dust-ion-acoustic solitary waves. The results are expected to be helpful in describing the rotating flows of magnetized plasma that are believed to exist in the rotating stars, the pulsar magnetosphere, and other rotating astronomical objects. We also explore the dynamic behaviors of multi-solitons as well as multi-shocks that propagate in a magnetized dusty plasma. The simplified Hirota method and the Cole-Hopf transformation are applied to construct the multi-soliton or multi-shock solutions. We observe that the magnetic field has a decreasing effect on the multi-soliton amplitudes and widths, whereas the dust concentration has an increasing effect on the amplitudes and widths of both types of waves. Most local government sites provide only informational and one way communications. All are still at the basic level of publishing information online. Two way interactive e-services have already been introduced with limited extent by some institutions in our country like online application of university admission, online job application to BPSC and Banks etc. Before implementing and developing any e-services, the authorities should know the expectation to the service from the users and it is also important to know the usability of the e-service in order to develop it with more usability function so that the users might satisfy by using the service. This study has attempted to assess the current standings of e-service(s) and what would be the prospective e-service(s) provided by BTCL. It has also endeavored to identify the barriers/challenges to adopt public e-service(s) in BTCL. Due to time and resource constraints only selective areas of Dhaka city has been surveyed. The study is based on both literature review and questionnaire survey. Two types of questionnaire were prepared. One for supply side i.e. for officials of BTCL and another was for demand side i.e. customer of BTCL. Questionnaire survey was conducted on total 52(fifty two) respondents. Of them, 27(twenty seven) was officials and the rest 25(twenty five) was subscribers of different areas of BTCL in Dhaka city. A literature review covers theoretical tools to define effective e-service and to identify hindrance in implementing e-service(s) in BTCL. The study has identified some prospective public e-services that BTCL should attain to retain its existence. It has also identified the possible barriers for implementing e-service(s). The perception of the officials and subscribers about prospective e-service(s) and hindering factors of e-service implementation is somewhat similar. The hindering factors are categorized in three types, organizational, procedural and technological. AbdusChronic subdural haematoma (CSDH) is a frequent neurological pathologic entities in daily neurosurgical practice with a rapidly rising incidence due to increasing age & wide spread use of anticoagulant. The "gold standard" of treatment is surgical evacuation by burr-hole craniostomy (BHC) for symptomatic patients but associated with complications, a recurrence rate of up to 30%, 1-year mortality rates could be as high as 32%. A complex intertwined pathway of inflammatory process, angiogenesis, local coagulopathy, recurrent microbleeds & exudates play a major role in the pathogenesis of CHDH. The objective of this study showed the effect of primary DXM therapy in treating CSDH is safe, beneficial, promising functional outcome & cost effectiveness as an alternative to BHC & surgical drainage in selected group of patients This study is a prospective, single centre, open labeled, randomized controlled clinical trial (RCT). Consecutive patients with a CSDH with MGS grade 1-3 will be randomized to treatment with DXM therapy or BHC. The DXM protocol will be 4 mg 8 hourly either oral or i/v for 21 days, which is then slowly tapered 1 mg per day every 3 days for 4 weeks.For insufficient haematoma resolution BHC can be performed.The primary outcomes are the functional outcome by means of the mRS score at 3 months & cost effectiveness at 12 months.Secondary outcomes are QOL at 3&12 months using the SF-36 & QOL/BRI, haematoma thickness after 2 weeks on follow-up CT, haematoma recurrence during the first 12 months,complications & drug related adverse effects,failure of therapy within 12 months after randomization & intervention,mortality during first 3&12 months,duration of hospital stay & overall healthcare & productivity costs. To test non-inferiority of DXM therapy Vs BHC,finally 30 patients in DXM therapy arm & 30 patients in BHC arm are required. The study started in June 2012; its outcomes demonstrates interesting alternatives to BHC in the management of patients harboring CSDH. Based on pathophysiologic mechanisms & patient studies treatment with steroid play a major role in the treatment of CSDH & as effective as BHC on functional outcome,at lower costs.Item SPATIO-TEMPORAL VARIATION OF SOME PHYSICO-CHEMICAL PARAMETERS AND ABUNDANCE OF PLANKTONIC COMMUNITY IN THE ATRAI RIVER, DINAJPUR, BANGLADESH(HAJEE MOHAMMOD DANESH SCIENCE AND TECHNOLOGY UNIVERSITY, DINAJPUR, 2019-06) Salam, Md. Rezoanul HaqueThe study was conducted to assess spatial and seasonal variations of some physico-chemical parameters and plankton community of the Atrai River in Dinajpur district for a period of one year from August 2017 to July 2018. The whole period was grouped into four seasons such as summer, autumn, winter and spring. Samples were collected monthly from three sampling sites named Vushi, Birga and Jhanjira assigned as site-1, site-2 and site-3, respectively. Some physico-chemical parameters namely, air and water temperature (°C), transparency (cm), depth (m), pH, dissolved oxygen (mg L-1 ) and alkanity (mgL-1 ) and plankton community were determined both at the field level and at the laboratory of the faculty of Fisheries, Hajee Mohammad Danesh Science and Technology University (HSTU), Dinajpur. The physicochemical parameters and plankton abundance did not vary significantly (P>0.05) among the sites but varied significantly (P<0.05) among seasons. However, all the physico-chemical parameters were within the acceptable limits for fishes, except transparency. The mean values of air temperature (°C)varied from 22.04±2.25 to 31.04±1.830C water temperature (°C)varied from 21.02± 2.29 to 30.24±1.990C and water transparency varied from 54.37± 9.98 to 67.74±13.58 cm. Water depth (m) was highest (6.66±1.34 m) in autumn andlowest (1.95± .17 m) in summer. The level of water of the river was influenced byseason. The pH value varied from 7.67±.53 to 9.42±.35 in autumn and winter, respectively. Dissolved oxygen concentration washighest (9.62±1.01 mg/L) in winter andlowest(8.69±.96 m/L) in autumn. The mean values of total alkalinity (mgL-1 ) varied from 29.18± 6.75 to 45.19± 2.90 mg/L in the present study. Total 28 phytoplankton genus identified of which 11 belongs to Chlorophyceae, 11 to Bacillariophyceae, 4 to Cyanophyceae and 2 to Euglenophyceae. Similarly, 9 genus of zooplankton were identified of which 3 belongs to Rotifera, 3 to Cladocera and 2 to Copepoda and 1 to Nauplius, the crustacea larvae. The number of genera and density of plankton showed sky-scraping seasonal variation during winter and summer. The overall density of plankton community were higher throughout the winter season.However, phytoplankton abundance was higher in winter and zooplankton in Autumn season. From this study, it can be concluded that physico-chemical parameters remain an acceptable range all the year but the plankton densities varies seasonally with higher in winter and lower in summer. This information could be helpful for further research work and fisheries management point of view.Item Volumetric and Viscometric Studies on the Long Chain fatty acid in Surfactant Stabilized Propanol(Khulna University of Engineering & Technology (KUET), Khulna, Bangladesh., 2015-01) Salam, Md. Abdus; Yousuf, Prof. Dr. Muhammad AbuVolumetric and viscometric studies of binary mixtures of (fatty acid + propanol) and ternary mixtures of (fatty acid + propanol + surfactant) were done. Lauric, palm tic and stearic acids were taken as fatty acids while the surfactant was sodium dodecyl sulfate (SDS). The investigated binary and ternary systems were carried out over a concentration range of 0.0100-0.4000 M of fatty acids at 298.15 to 323.15 K at 5 K intervals. The SDS concentrations were 0.001 M and 0.005 M. The apparent molar volumes, φv. of both binary and ternary mixtures were determined from the densities of the mixtures. The φv, values have been found to be positive throughout the whole concentration range for all acids in propanol. The apparent molar volumes, .co. of all fatty acids in propanol were found to be increased with concentration at all temperatures.SimilarlyφV, values were found to be increased with increasing temperature at any concentration of the solution. These results may be due to the solute—solvent and solute—solute interaction in the solutions. This shows that φ, values are strongly dependent upon concentration as well as the temperature. The φv values are also found to be increased with the chain length of the fatty acids and this may be due to the hydrophobic characters of alkyl chain. The number of carbon atoms in the hydrocarbon chain of the fatty acid is proportional to the degree of hydrophobic/hydroPhilic balance of the whole compound. Eventually the pattern of apparent molar volumes of laurie, palmitic and stearic acids in 0.001 and 0.005 M SDS solution is very similar to those of (propanol + fatty acid) systems. The apparent molar volumes decreased at all concentration of SDS in comparison to corresponding systems without SDS. The results conferred that at molecular arrangements in SDS to (fatty acid+propanol) systems more organized than corresponding systems without SDS. The values of viscosity (η) of laurie, palmitic, and stearic acids in propanol increased with concentration at all six temperatures. The increase of η values of all acids with concentration can be attributed to the increase in both solute—solvent and solute—solute interactions with 17 are found to be increased with the carbon number along the hydrocarbon chain of the fatty acid concentration. At the same time, at a constant temperature and concentration, values of η. can be explained by the increase in both molecular mass of the fatty acid and the degree of solute solvent interactions with carbon number. The ultimate pattern of viscosity after addition of SDS is almost similar to corresponding systems without SDS. The change of free energy, ∆G and enthalpy, ∆H* values for viscous flow were found to be positive work had to be done to overcome the energy barrier for the flow process. The change of entropy, ∆S* values for the flow process are negative in all cases.
