Item EFFECT OF MICRONUTRIENT ZINC ON THE PERFORMANCE OF MODERN RICE VARIETIES(DEPARTMENT OF SOIL SCIENCE, 2021) SULTANA, RAZIAThe experiment was carried out during the period from December 2020 to May 2021 at the research field of the Bangladesh Rice Research Institute, Regional Station, Sonagazi, Feni, Bangladesh. The experiment consisted of two factors: Factor A: eight Zn levels viz. T 1 (Recommended rate: N, P, K, S and Zn (monohydrate) @ 124, 22, 75, 20 and 4 kg ha ), T 2 (T 1 – 50% Zn), T 3 (T 1 – 100% Zn; T 1 but without Zn), T 4 (T 1 + 50% higher Zn; ZnSO 4 : 13.20 kg ha -1 ), T 5 (T 1 + 100% higher Zn; ZnSO 4 : 15.40 kg ha -1 ), T 6 (T 1 but Zn was applied by spraying), T 7 (Soil test based (STB) nutrient rates N, P, K, S and Zn @ 158, 24, 82, 10 and 2 kg ha -1 ) and control treatment T 8 (control/native supply of nutrient; no nutrient was applied) and Factor B: three rice varieties viz. V 1 (BRRI dhan74), V 2 (BRRI dhan84) and V 3 (BRRI dhan88). The experiment was laid out in Strip Plot Design with three replications. Among different levels of micronutrient zinc (Zn) treatments, the longest plant height (90.4 cm), maximum number of tillers m -2 (472), panicles hill -1 (11.6), and panicles m -2 (385); and highest grain yield (5.28 t ha -1 ) were recorded in the treatment T 5 + 100% higher Zn; ZnSO 4 :15.4 kg ha -1 ) and this treatment also showed highest Zn content in grain (0.136 ppm), however the highest Zn content in straw (0.403 ppm) was acquired from T 6 (T 1 but Zn was applied by spraying). Again, the treatment T 1 (Recommended rate: N, P, K, S and Zn @ 124, 22, 75, 20 and 4 kg ha -1 1 (988) and 1000-grain weight (23.4 g). Control treatment T 8 ) gave the highest number of grains hill (no nutrient was applied) gave least performance for the respected parameters. Among three rice varieties, the highest number of tillers m -2 (446) and grains hill -1 (920) were achieved by V 3 (BRRI dhan88), while the maximum number of panicles m -2 (369) and the highest mean grain yield (4.56 t ha -1 ) were produced by V 1 (BRRI dhan74). Moreover, different varieties showed nonsignificant variation on Zn content in grain and straw. Considering the combined effect of Zn treatments and variety, the highest number of panicles m -2 (425), 100-grain weight (27.1 g) and highest grain yield (5.73 t ha -1 ) were observed in T 5 V 1 . In case of Zn content of grain and straw, the maximum (0.218 and 0.573 ppm, respectively) were found in T 5 and T 6 V 3 , respectively, whereas T 8 V 2 gave the minimum Zn content in grain and straw (0.046 and 0.088 ppm, respectively). Based on overall performances, it could be concluded that the treatment T 5 (T 1 + 100% higher Zn; ZnSO 4 : 15.40 kg ha -1 ) might be recommended for sustainable cultivation of modern rice varieties to obtain higher yield and better Zinc content.Item IN VITRO REGENERATION OF ORCHID (Dendrobium sp.)(DEPARTMENT OF BIOTECHNOLOGY, 2021) SULTANA, RAZIAThe experiment was conducted at the Biotechnology Laboratory of Department of Biotechnology, Sher-e-Bangla Agricultural University, Dhaka from January 2021 to June 2021 to investigate the effect of different plant growth hormone for shoot proliferation and root regeneration of Dendrobium sp. where shoot tips were used as explant. The experimental data were taken on different parameters of shoot and root growth. Different doses of individual shooting hormone(BA), individual rooting hormone(NAA) and the combination of these two hormones were experimented here. The minimum days to shoot induction were recorded 15 days from treatment T 8 (2.0 mg/L BA + 1.50 mg/L NAA). Maximum percentage (73.4%) of shoot induction was noticed in treatment T 8 (2.0 mg/L BA + 1.50 mg/L NAA) .The maximum days to root induction was 44 days and it was recorded in control treatment .The minimum days to root induction was 17 and it was observed from treatment T 8 (2.0 mg/L BA + 1.50 mg/L NAA) days. In the following study significant variation was observed for the use of BA and NAA on the percent of shoot induction explants. Significant variation was observed for the use of BA and NAA on the percent of root induction of explants . The treatment of 2.0 mg/L BA response excellent in shoot induction and 1.0 mg/L NAA showed better performance in root development in Dendrobium sp. Combined effect of BA and NAA was very significant. Overall a reliable protocol was developed for shoot and root proliferation which can be used for further crop improvement program.Item STUDY ON EFFECT OF STORAGE CONDITIONS ON HEALTH STATUS OF OKRA SEEDS(Department of Plant Pathology, 2013-12) SULTANA, RAZIAAn experiment was conducted in Seed Pathology Lab in the Department of Plant Pathology, SAU, Sher-e-Bangla Nagar, Dhaka-1207 during July 2013 to June 2014 to determine the quality and health of okra seed and determination of seed health status during seed storage. Five storage container viz. T 1 : plastic container, T : plastic bag, T 3 : polythene coated gunny bag, T 4 : gunny bag and T : earthen pot and three seed treatments viz. N 0 : no treatment, N 1 5 : seed treated with Provax-200 and N : seed treated with neem leaf powder extract were tested. Data were recorded on the percent infestation of storage seeds by fungi and also on quality attributes under different treatments during the seed storage at 4, 8 and 12 months. The experiment was laid out in two factors Completely Randomized Design (CRD) with three replications and the mean were adjusted by DMRT at 5% level. In case of storage container and seed treatments, provax-200 treated seeds stored in plastic container (T 2 1 N ) showed significantly lower moisture content (7.98%) at 4 months of storage than that of other treatments while non - treated seeds stored in gunny bag (T 1 ) showed the maximum moisture content (17.66%) at 12 months of storage. Similarly, percentage of seed infested minimum in T 1 N The prevalence of different fungi were Fusarium spp (0.13%), Chaetomium globosum (0.72%), Aspergillus flavus (0.52%), Aspergillus niger (0.87%), Rhizopus stolonifer (0.59%) and Curvularia spp (0.80%) at 4 months of storage. Seed infested was highest in T 4 N 0 1. where prevalence of Fusarium spp (9.27%), Chaetomium globosum (11.96%), Aspergillus flavus (18.26%), Aspergillus niger (16.63%), Rhizopus stolonifer (8.72%) and Curvularia spp (8.20%) at 12 months of storage. In contrast, provax200 treated seeds stored in plastic container (T 1 N ) showed the maximum germination (96.74%), longest shoot (21.04 cm) and root (11.91 cm), highest seed 1 4 N 2 0 vigor index (3229.0), highest weight of dry seedlings (55.72 g) at 4 months of storage while non-treated seeds stored in gunny bag (T 4 N ) showed lower performance in seed germination (39.71%), shoot length (11.33 cm), root length (6.20 cm), seedling vigor index (731.30), dry weight of seedlings (33.33 g) and filed emergence (34.51%) at 12 months of storage in this study. The above result proved that the studied attributes considerably increase with increasing storage period except seed germination, seedlings shoot and root length, seed vigor index, dry weight of seedlings and field emergence where plastic container (T 0 ) and seed treated with provax-200 (N ) singly or their interaction effect had highly efficient for obtaining the greater performance regarding quality attributes of storage okra seed and also for controlling the fungi species. The storage plastic container along with provax-200 treated seeds showed the best storage performance in storage of okra seed. 1 1
