EkramulThe research study was conducted to determine the antitumor effect of the flower of Alangium salvifolium (crude extract and diethylether fractions) against Ehrlich Ascites Carcinoma (EAC) in mice at the doses of 10 mg kg-1 body weight intraperitoneally. Extract/fractions was administered for nine consecutive days. Twenty-four hours of last dose and 18 h of fasting, the mice were sacrificed and antitumor effect was assessed by evaluating tumor volume, viable and nonviable tumor cell count, tumor weight and hematological parameters of EAC bearing host. Significant (p<0.001) increases of survival times 30±0.96 and 25±0.40 days for crude extract and diethylether fraction of the A. salvifolium (10 mg kg-1) treated tumor bearing mice, respectively were confirmed with respect to the control group (20±0.13 days). The extract/fraction also decreased the body weight of the EAC tumor bearing mice. Hematological studies reveal that the heamoglobin (Hb) content was decreased in EAC treated mice whereas restoration to near normal levels was observed in extract treated animals. There was a significant (p<0.001) decrease in RBC count and increase in WBC counts in extract/fraction treated animals when compared to EAC treated animals. Antioxidant potential of the EA extract was evaluated in vitro by DPPH (1,l-diphenyl-2-picrylhydrazyl) scavenging assay method. The EA extract showed free radical scavenging activity with IC50 value of 24.08 ± 0.30 fig/mL. The antitumor activity of the extract was determined against Ehrlich ascites carcinoma (EAC) in mice at 25 mg/kg and 5Í) mg/kg body weight intraperitoneal. Significant (p<0.001) increase of survival time by 25 ± 0.57 and 27 ± 0.40 days by the EA extract treated tumor bearing mice was confirmed with respect to the control group (22 ± 0.12 days). Hematological studies revealed that the hemoglobin (Hb) content was decreased in EAC treated mice whereas restoration to close to normal levels was observed in extract treated animals. There was a significant (p<0.001) decrease in RBC and increase in WBC counts in extract treated animals when compared to EAC affected animals. Therefore, the EA extract of P. simiarum was capable to exhibit moderate antioxidant and antitumor activities. Therefore, this study is designed to investigate its antioxidant, analgesic and CNS depressant effects. Materials and Methods: The antioxidant activity of methanolic extract of S. nodiflora (SN) was determined using Folin Ciocalteu reagent, phosphomolybdenum method, free radical scavenging activity by 1,1-diphenyl-2-picryl-hydrazyl (DPPH) and the reducing power activity. Analgesic activity was done by acetic acid and formalin model and CNS depressant activity was measured by the hole cross and open field method. Results: The total phenols and total antioxidant capacity of SN was found to be 37.38 < 2.01 mg/g equivalent of gallic acid and 433.37 < 4.22 mg/gm equivalent of ascorbic acid. The percentage (%) scavenging of DPPH free radical was found to be concentration dependent and IC50 value was 125.89 < 2.23 µg/ml while the IC50 value of standard ascorbic acid was found to be 16.76 < 0.11 µg/ml. The reducing power of SN was found to be concentration dependent. Acetic acid and formalin models are peripherally acting analgesic methods. The oral administration of both doses (100 and 200 mg/kg b.wt.) of SN significantly (p<0.001) inhibited 40.06% and 61.83% writhing response induced by acetic acid whereas oral administration of the same doses of SN significantly (p<0.001) inhibited 56.96% and 62.60% itching response induced by formalin. The methanolic extract of SN, at the dose of 250 mg/kg and 500 mg/kg b.wt. produced significant (P<0.001) decrease of locomotion. Glucose control plays an important role in the pro-oxidant/antioxidant balance. Some antidiabetic agents may by themselves have antioxidant properties independently of their role on glucose control. Objective: The present investigation draws a comparison of the protective antioxidant activity, total phenol content and the antihyperglycemic activity of the methanolic extract of Cajanus cajan root (MCC) and Tamarindus indica seeds (MTI). Materials and Methods: Antidiabetic potentials of the plant extracts were evaluated in alloxan-induced diabetic Swiss albino mice. The plant extracts at the doses of 200 and 400 mg/kg body weight was orally administered for glucose tolerance test during 1-hour study and hypoglycemic effect during 5-day study period in comparison with reference drug Metformin HCl (50 mg/kg). In vitro antioxidant potential of MCC and MTI was investigated by using 1, 1- diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity at 517 nm. Total phenolic content, total antioxidant capacity and reducing power activity was also assayed. Results: There was a significant decrease in fasting serum glucose level (P < 0.001), reduction in blood glucose level (P < 0.001) in 5-days study, observed in the alloxan-induced diabetic mice. The reduction efficacy of blood glucose level of both the extracts is proportional to their dose but MCC is more potent than MTI. Antioxidant study and quantification of phenolic compound of both the extracts revealed that they have high antioxidant capacity. Conclusion: These studies showed that MCC and MTI have both hypoglycemic and antioxidant potential but MCC is more potent than MTI. Results: There was a significant decrease in fasting serum glucose level (P < 0.001), reduction in blood glucose level (P < 0.001) in 5-days study, observed in the alloxan-induced diabetic mice. The reduction efficacy of blood glucose level of both the extracts is proportional to their dose but MCC is more potent than MTI. Antioxidant study and quantification of phenolic compound of both the extracts revealed that they have high antioxidant capacity. Conclusion: These studies showed that MCC and MTI have both hypoglycemic and antioxidant potential but MCC is more potent than MTI. The present study suggests that both MCC and MTI could be used in managing oxidative stress.Item Correlation Between Heavy Metals and Neonatal Hyperbilirubinemia Among the Patients Attended at Bangabandhu Sheikh Mujib Medical University(University of Rajshahi, 2020) Hossain, Mohammad Anwar; Islam, Md.Ekramul; Mosaddik, Ashik; Islam, Md. SaifulNeonatal hyperbilirubinemia is a frequently encountered problem in 60%–80% of newborns worldwide. The burden is high in low-income and middle-income countries like Bangladesh. A recent report noted that at least 4,81,000 term/near-term neonates are affected by hyperbilirubinemia each year, with 1,14,000 dying and an additional 63,000 surviving with kernicterus. Under this circumstance, it is essential to tackle severe neonatal jaundice as one key component of optimizing neurodevelopmental outcome to achieve the sustainable development goals. Therefore, this study has designed to find the incidence and relation of neonatal jaundice with heavy metals such as copper, zinc and magnesium to inform child health policy regarding its prevention and management. About 594 neonates were collected from the outpatient and inpatient Department of Clinical Pathology (Laboratory medicine) and Neonatology at Bangabandhu Sheikh Mujib Medical University (BSMMU), Dhaka. Infants with severe congenital malformation, sepsis, or birth asphyxia were excluded from this study. They were divided into case and control groups. This study was approved by the institutional ethical committees of Institute of Biological Sciences (IBSc), University of Rajshahi, Bangladesh (Approval Memo no-82/320/IAMEBBC/IBSc, 20 August, 2017). The mean age of the respondents was 3.08 and 3.21 days; the sex variable of the respondents were male 187 and 160 in case and control group respectively and female were 132 and 115 in case and control group respectively which was not significant. The mean weight of the respondents was 2.62±0.4 and 2.84±0.3 kilogram which was significant. The concentration of serum bilirubin was significantly (p<0.001) higher (171.14 mg/L) in case group when compare to control group (35.42 mg/L). Because the liver function becomes progressively worse in case group. The serum magnesium was 23.98 mg/L and 19.19 mg/L in case and control group, respectively. The concentration of serum magnesium is higher in case group when compared with control which is very significant (p<0.001). The level of serum zinc was 0.50 mg/L in case group and 0.68 mg/L in control group. The serum zinc level is significantly (p<0.001) lower in case group when compared with control group. In case group the level of copper was 0.75 mg/L and in control group was 0.43 mg/L. It was significantly (p<0.001) higher in case group when compared with control group. There was a positive significant Pearson correlation (r=0.817, pvalue<0.001) between serum total bilirubin level and serum magnesium level in both groups. It means if serum bilirubin is increased, serum magnesium also increased in case group and control group. Relation between serum total bilirubin level and serum zinc level was negative significant Pearson correlation (r= -0.773, p value <0.001) in both groups. It is justified that if serum bilirubin level is increased the serum zinc level also decreased in case group and control group. The positive significant Pearson correlation (r=0.832, p value <0.001) was in between serum total bilirubin and serum copper level in both groups and it can be said that if serum bilirubin level is increased, the serum copper level also increased in case group and control group. So, in this study, the heavy metal like zinc, copper and magnesium has relation with neonatal hyperbilirubinemia. If serum bilirubin is increased, serum magnesium and copper also increased but when serum bilirubin level is increased, the serum zinc level decreased in the patient of neonatal hyperbilirubinemia.Item Free radical-scavenging activities and cytoprotective effect of polyphenol-rich ethyl acetate fraction of guava (Psidium cattleianum) leaves on H2O2-treated HepG2 cell(© 2013 Springer Link, 2013-12) Mosaddik, Ashik; Moon, Jeong Yong; Lee, Sarah; Jeong, Seongweon; Kim, Jong-Chan; Ahn, Kwang Seok; Cho, Somi KimTotal phenolic contents of different fractions of the Psidium cattleianum leaf extract and their antioxidant capacity against several free radicals were examined. Protective effect of the ethyl acetate fraction (EAF) on H2O2-induced DNA damage in HepG2 cells were also evaluated, and the phytochemical profile of EAF was analyzed using tandem mass spectrometry. EAF derived from the 80% methanol extract of the leaf contained a remarkable amount of polyphenol and showed high levels of DPPH and alkyl radical scavenging activity, promoted cell viability, and protected against H2O2-induced DNA damage in HepG2 cells. Phytochemical analysis revealed that the major components in the EAF included quercetin monoglycoside, phloridizin, quercetin 3-diglycoside, quercetin-3-glucuronide, 2,6-dihydroxy-3,5-dimethyl-4-glucopyranosylbenzophenone, phenolic acid, guaijaverin, and naringin. The present study suggests possible synergistic or competitive antioxidant action of the major compounds of cattley guava leaf on H2O2-induced DNA damage in HepG2 cells.
