    ANTIMICROBIAL CONSTITUENTS FROM INDIGENOUS PLANTS GROWING IN BANGLADESH

    This thesis report about the separation and structure explanation of secondary metabolite from the indigenous plant named Stereospermum suaveolens (Roxb.) DC(family-Bignoniaceae). A total eight (08) compounds were isolated of which one (SSC-003, Figure: 3.9) appears to be new. The compositions of the separated compounds were described mainly by NMR and mass spectroscopic techniques besides, Stereospermum suaveolens (Roxb.) DC and Diospyros malabarica (Desr.) Kostel (Family-Ebenaceae) buds was investigate for biological (antimicrobial, antioxidant, cytotoxic, thrombolytic, membrane stabilizing, analgesic, anti-diarrheal, CNS antidepressant and anti-diabetic) activity studies of the extractive from the plant. Repeated chromatographic isolation and purification of the n-hexane soluble part (HESF) of methanolic crude of the leaves and steam bark of S. suaveolens afforded two (02) compounds SSC-001 (Stigmasterol, Figure: 3.1) and SSC-002 (β-amyrin, Figure: 3.6). Between these two compounds SSC-002 (β-amyrin, Figure: 3.6) is separated for the first time from this plant. The carbon tetrachloride soluble part (CTSF) of leaves and steam bark of S. suaveolens yielded six (06) compounds designated as SSC-003 (19,23-dihydroxy-3-oxa-1-ursane, Figure: 3.9),SSC-004 (Uncaric acid, IUPAC name: 3β,6β,19α trihydroxyurs 12 ene 28 oic acid, Figure: 3.20), SSC-005 (Oleanolic acid Figure: 3.30), SSC-006 (p-Methoxy-trans-cinnamic acid, Figure: 3.35), SSC-007 (p-Methoxy-cis-cinnamic acid, Figure: 3.40) and SSC-008 (Ilexolic acid B, IUPAC name: 3β,19α dihydroxyolean 12 ene 23,28 dioic acid, Figure: 3.49). Compounds SSC-006 (p-Methoxy-trans-cinnamic acid, Figure: 3.35) and SSC-007 (p-Methoxy-cis-cinnamic acid, Figure: 3.40) have been also parted from chloroform soluble fraction (CLSF) of leaves and bark of S. suaveolens. Among these six compounds, SSC-003 (19,23-dihydroxy-3-oxa-1-ursane, Figure: 3.9) represents a 30-carbon containing triterpenoid which appears to be new. Rests of these five compounds are firstly isolated from this genus and species. The methanol extraction of S. suaveolens was researched for analgesic, antidiabetic, anti-diarrheal and anti-depressant activity on Swiss Albino mice. Antidiabetic activity was take measure by orally feed glucose tolerance test as the methanolic crude of S. suaveolens at 200- and 400- mg/kg b.w exhibited 37.58% and 56.10% decreased of blood glucose level as II along with 58.53% by standard glibenclamide (3 mg/kg b.w.) after 120 min induced the glucose. Castor oil-induced diarrhea in mice, the plant methanolic extract of 200- and 400- mg/kg b.w. demonstrated 23.08% and 46.15% while the standard loperamide (50 mg/kg b.w) exhibited 57.89% decrease of diarrheal feces. In tail flick method in mice for central analgesic activity, the methanolic extract dose of 200- and 400- mg/kg b.w. exhibited highest elongation (185.59% and 259.21% respectively) compared to 313.32% by morphine (4 mg/kg b.w) after 60 min. In peripheral analgesic activity assessment, the test samples, doses of 200- and 400- mg/kg b.w. displayed 31.82% and 48.48% reduction of writhing respectively as compared to 65.15% inhibition reduced by the standard diclofenac-Na (50 mg/kg b.w). In evaluation of anti-depressant activity, the crude of S. suaveolens (200- and 400-mg/kg b.w) didn’t show any remarkable CNS anti-depressant activity. The methanolic crude and its different Kupchan fragments of S. suaveolens were also based on screenings for total phenolic content, DPPH free radical scavenging assay, brine shrimp lethality bioassay (Cytotoxicity), membrane stabilizing, thrombolytic and antimicrobial activities. In antioxidant activity, aqueous soluble fraction of Kupchan partition showed highest IC50 value of 18.99μg/ml and highest phenolic content was found also (9.7 mg of GAE/gm of dried extract). Membrane stabilizing activity was assessed by hypotonic solution- and heat-induced methods and was compared to reference acetyl salicylic acid. In hypotonic solution-induced hemolysis, the n-hexane and the carbon tetrachloride soluble parts demonstrated 54.42% and 52.67% inhibition of hemolysis of RBCs. In heat-induced hemolysis, the chloroform soluble fragments revealed the hemolysis of RBC by 57.10% as compared to 72.09% produced by acetyl salicylic acid. In brine shrimp lethality bioassay, there was no remarkable lethality (IC50) observed. In the antimicrobial properties determination by disc diffusion method, only the n-hexane and carbon tetrachloride soluble parts revealed moderate activity (inhibition = 7.0-15.0 mm) against the test organisms. It is evaluated that the methanolic crude of S. suaveolens contains chemically unique and diversified chemical constituents. The plant extractives exhibited significant analgesic, anti-diarrheal and ant anti-hyperglycemic activities but moderate antioxidant, membrane stabilizing and thrombolytic properties. However, the test sample demonstrated weak antimicrobial activities. III The other part of present thesis was planned to assess the antioxidant, antimicrobial, antidiarrheal and analgesic activities of methanol crude and its different kapchan partitions of Diospyros malabarica (Desr.) Kostel buds growing in Bangladesh. In evaluation of antimicrobial assay, all the test samples showed prominent antimicrobial activity against the experimental organisms under in vitro conditions. Among these, the carbon tetrachloride soluble fragments exhibited the highest activity ranging 20-32mm in diameter. In antioxidant activity, aqueous soluble fraction of Kupchan partition showed highest IC50 value was 6.17 for DPPH assay test and also exhibited highest phenolic content was 12.87 mg of GAE/gm Gallic acid equivalence. In assessment of antidiarrheal activity, the D. malabarica extracts displayed significant anti-diarrheal potential in a dose dependent manner, the dose of 200- and 400- mg/kg b.w. displayed 26.92% and 50.00% while the standard loperamide (50 mg/kg b.w) revealed 53.85% reduction of diarrheal feces. During the assessment of analgesic activity by radiant heat tail-flick method, the plant sample at 400 mg/kg b.w. exhibited highest elongation (373.04%) as compared to morphine (435.88%) after 90 minutes of administration. In acetic acid-induced writhing test, the extract at 200- and 400-mg/kg b.w. showed 61.11 and 66.67% reduction of writhing in mice model, respectively as compared to 68.06% inhibited by the standard diclofenac-Na. The findings of this study justify some of the traditional uses of D. malabarica and reveal the bioactivity of the plants. Further studies are required to isolate and identify the bioactive compounds.

    EFFECT OF NITROGEN AND SULPHUR ON THE GROWTH AND YIELD OF MAIZE

    A field experiment was carried out at the research field of Crop Physiology and Ecology department, Hajee Mohammad Danesh Science and Technology University, Dinajpur during April to July 2018 to evaluate the performance of maize (BARI hybrid Bhutta-7) as influenced by different levels of nitrogen and sulphur. The experiment comprised of two factors; such as factor A: Three nitrogen levels, N0: 0 kg N/ha (control), N1: 100 kg N/ha, N2: 150 kg N/ha and factor B: four levels of sulphur like, S0: 0 kg S/ha, S1: 15 kg S/ha, 25 kg S/ha and S3: 35 S/ha. The experiment was set up in a randomized complete block design with three replications. Both nitrogen and sulphur significantly affected growth, yield and yield contributing characters of maize. In case of nitrogen, the tallest plant, highest leaf area, branch number per plant, maximum days to tasseling, silking and maturity were observed in N2 (150 kg N/ha) but leaf number and plant dry weight was highest in N1 (100 kg N/ha) and the result was minimum for all above parameters in N0 (0 kg N/ha). In case of sulphur, the tallest plant, highest leaf area, leaf number, branch number per plant, plant dry weight, maximum days to tasseling, silking and maturity were observed in S2 (25 kg S/ha) and the result was tolerant for all above parameters in S0 (0 kg S/ha). In interaction, the tallest plant, highest leaf area, branch number per plant, maximum days to tasseling, silking and maturity were observed in N2S2 interaction but leaf number and plant dry weight was highest in N1S2 interaction whereas the result was lowest for all above parameters in N0S0 interaction. In case of yield and yield contributing characters like cob length, number of cob per plant, number of grains/cob, 1000-grain weight, grain yield and stover yield had significant variation due to different nitrogen and sulphur levels. In different nitrogen levels, the best performance was observed in N1 (100 kg N/ha) whereas, the lowest performance was observed in N0 (0 kg N/ha). In different sulphur levels, best performance observed in S2 (25 kg S/ha) and the lowest performance was observed in S0 (0 kg S/ha). Therefore in interaction, highest cob length, number of cob per plant, number of grains/cob, 1000-grain weight, grain yield and stover yield were found in N1S2 interaction and the result was opposite for all above parameters in N0S0 interaction. Based on the above findings it can be concluded that an application of 100 kg N ha-1 with 25 kg of S ha-1 is the best combination of nitrogen and sulphur for obtaining higher yield attributes and yield of hybrid maize.

    EFFECT OF SOWING DATE AND IPRODIONE ON THE ALTERNARIA BLIGHT OF MUSTARD (Brassica juncea)

    A field study had been carried out during the period from October 2006 to February 2007 at Syedpur in Nilphamari district to determine the effect of sowing date and Iprodione in controlling the Alternaria blight of mustard cv. Tori 7. The three sowing dates were 23 October (early), 07 November (normal) and 22 November (late), 2006. Iprodione (0.25%) was applied three times, viz. single spray, two sprays and three sprays at 10-days interval starting from the first disease initiation on the lower older leaves of 30-40 days old plants. Control plots were sprayed with plain water. The manipulation of early sowing date as non-monetary input decreased the leaf area diseased, pod area diseased, number of pod infection, number of spot per pod and increased 1000-seed weight, seed yield per plant and yield per hectare over late sowing. Results obtained in this study indicated a tremendous effect of sowing date on the disease severity and seed yield of mustard. The application of three sprays of Iprodione provided the maximum control of Alternaria blight and increased seed yield per hectare. Iprodione three sprays in combination with early sowing (23 October) displayed the best performance in controlling the Alternaria blight disease. Three sprays of Iprodione (0.25%) or 23 October (early) sowing alone significantly brought down the disease severity while in combination reduced the disease significantly and increased seed yield by 28.42 % over control (non-sprayed) of late sowing crop. Finally, it may be concluded that Iprodione three sprays in combination with early sowing was found superior for management of Alternaria blight of mustard oilseed crop.

    FACTORS AFFECTING ON THE CONCEPTION RATE OF DAIRY CATTLE AT FIRST SERVICE IN CHAR AREAS OF NORTHERN BANGLADESH

    The study was conducted on the char areas of eight districts in namely Bogra, Sirajgonj, Pabna, Jamalpur, Kurigram, Nilphamari, Rangpur, Gaibandha, respectively. However, data was collected from January, 2012 to December, 2013 by the structured questionnaires, informal interview and group discussion. A total of 800 local, 800 Holstein-Frisian (L×HF) and 800 Red Chittagong (L×RC) crossbred cows were selected for this study. Feeding and management of the experimental animals were uniform throughout the year. Animals were vaccinated and dewormed regularly. Cows generally were grazed in the field and supplied 5−6 kg straw daily at evening. In improved feeding system cows were grazed in the field and supplied ready feed (a mixture of rice polish, wheat bran, broken rice and oil cake; @ 2–3.5 kg/d/head) with 5−6 kg straw. The cows accessed ad libitum fresh drinking water with iodized salt. The results revealed that though first service conception rate was not significantly influenced by cows genotypes, but it was significantly influenced by bull genotypes when the RC crossbred cows inseminated naturally by local bull (73%) than the cows inseminated (AI) by the semen of L×HF crossbred bull (61%). The cows showed moderate conception rate inseminated naturally by L×RC crossbred bull (66%). The conception rate of cows is influenced by the quality and quantity of semen, it was higher in the cows inseminated by using the semen supplied by Department of Livestock Services as compared to the semen supplied by the Ejab, even though the semen of BRAC. The variations in conception rate were actually due to the ultimate variability of semen quality which were depends on bull genotype, semen collection technique, processing, preservation, heat detection, experience of AI technicians, etc. Farmers who use professional Al technicians from reputable Al organizations usually have fewer concerns about semen quality being a major cause of low conception rates. It was higher in the cows inseminated in spring season than the cows were inseminated specially in summer season. It was significantly varied on the experience of AI technicians. The conception rate was significantly higher in the cows were 2.5−3.0 years old (68 %) than the cows were 4.5−5.0 years old (54%). Conception rates were significantly higher in the cows provided improved diet, and managed under regular vaccination and deworming practices. Therefore, it may be suggested that to achieve the satisfactory conception rate in char areas, the farmers should inseminate their cows during spring season with experience AI technician by using good quality semen. They may choose to rear Red Chittagong cows or their crossbreds. They should provide concentrate feed at least a minimum amount and have to practice of regular vaccination and deworming to their cattle.

    VARIABILITY STUDY IN F2 MATERIALS OF INTERVARIETAL CROSS OF BRINJAL (Solanum melongena L.)

    An experiment was conducted to study genetic variability, character association and path coefficient analysis for yield and its contributing traits in Brinjal genotypes. The experiment was carried out at the Ilorticulture farm of Shre-E Bangla Agriculture University. Shre-E Bangla Nagar Dhaka-1207. The results indicated that diiTeren brinjal genotypes differed significantly regarding all the characters studied. Minimum differences of genotypic and phenotypic variances and coefficient of variation as well as high heritability coupled with high genetic advance and genetic advanced in percent of mean were observed for all the traits of different brinjat genotypes indicated additive gene effects of these traits. Number of fruit per plant and single fruit weight of different brinjal genotypes had a high degree of significant positive association with yield per plant and high positive direct effect indicated that these characters had the major contribution towards the yield per plant. Selection could be made based on these two traits for the improvement of yield of briqjal genotypes. Regarding singal fruit weight and yield per plant. the genotype Line-Ol x Line-25F2 perthrmed best. Genotypes Line-Ol x Line-25F1, I,ine-23x Line-24F2 produced higher yields next to genotype l,ine-Olx Line-251'- 2. In respect of days to flowering and days to first harvest Line_23x Linc-24F'2 was Ibund as the earliest genotype.
