    Biological Assessment, GC-MS Analysis, and Molecular Docking Investigation on the Neuropharmacological, Anti-Diarrhoeal, and Cytotoxic Properties of Ficus semicordata Fruits

    Background: Ficus semicordata Buch. is a well-known ethnomedicinal plant that is used to treat various ailments such as colic pain, urogenital difficulties, gastrointestinal disorders, visceral blockage, leprosy, jaundice, diabetes, and hepatitis. This study aims to investigate the phytochemical contents of the metabolites extracted (methanol) from the fruits of Ficus (F.) semicordata, and determine their neuropharmacological, anti-diarrhoeal, and cytotoxic potencies, using in vivo, in vitro, and in silico methods. Methods: The pharmacological properties of methanol extract of Ficus semicordata fruits (MEFSF) were assessed at different concentrations and its toxicity was determined using the in vitro brine shrimp lethality test. Tail suspension and forced swimming tests were used to investigate the antidepressant activity of MEFSF in mice and elevated plus maze and hole board test models were used to uncover its anxiolytic potentiality. The in vivo anti-diarrhoeal properties of MEFSF were tested on castor oil-induced diarrhoea and gastrointestinal motility models. Gas chromatography-mass spectrometry (GC-MS) analysis was conducted using a mass spectrometer. Based on the GC-MS analysis, 19 phytochemicals were investigated using molecular docking techniques against various target proteins to determine whether they mediate cytotoxic, depressive, anxiolytic, and anti-diarrhoeal effects. Results: MEFSF exhibited moderate toxicity (median lethal dose (LD50): 267.23 μg/mL). In the antidepressant assessment, MEFSF demonstrated a significant (p < 0.0001) dose-dependent decrease in immobility compared to fluoxetine. Similarly, MEFSF exhibited a dose-dependent reduction in anxiolytic-like behaviour in mice, with a 400 mg/kg dose exhibiting vigorous activity. MEFSF also significantly inhibited motility in both anti-diarrhoeal models, with a 400 mg/kg dose exhibiting highly significant (p < 0.0001) suppression. The GC-MS analysis revealed 81 bioactive components. Seven phytochemicals exhibited a strong affinity for various target proteins in the molecular investigation. Notably, beta-D-glucopyranose and 4-O-beta-D-galactopyranosyl manifested a high affinity for hER, K+ channel, SERT3, and M3MAR and exhibited cytotoxic, anxiolytic, antidepressant, and anti-diarrhoeal potential. Conclusion: The findings indicate that MEFSF can potentially contribute to the development of innovative anti-cancer, neuropharmacological, and anti-diarrhoeal treatments. However, additional research is necessary to explore this possibility.

    Biological investigations of the methanol extract of Tetrastigma leucostaphylum (Dennst.) Alston ex Mabb. (Vitaceae): In vivo and in vitro approach

    Tetrastigma leucostaphylum (Family: Vitaceae) is popular for its medicinal value in Bangladeshi tribal communities. This study aims to investigate several pharmacological values of methanol extract of T. leucostaphylum (METL). In vivo analgesic and anti-inflammatory researches have been implemented by using acetic acid-induced writhing and formalin-induced paw licking test protocols in mice. Furthermore, in vitro thrombolytic and anthelmintic studies have been performed by following the blood clot lysis method and nematode mortality measurement method. In the in vivo study, METL did not minimize the acetic acid-induced writhes prominently but significantly attenuate both the peripheral and inflammatory pain in mice in a dose-dependent manner. In early and late phase, METL 400 (mg/kg, b.w; p.o) showed 39.63 % and 48.73 % paw licking inhibition. Again, METL (100 μL) reflected 56.62 % clot lysis in thrombolytic research. Besides, METL causes the death of nematodes in a dose-dependent manner. The bioassay of the methanol extract of T. leucostaphylum justified the analgesic, anti-inflammatory, thrombolytic and anthelmintic activities of the crude extract and finally suggests the test extract as a wellspring of anti-inflammatory, thrombolytic and anthelmintic agents as a crude drug source.

    Chemical Profiles and Pharmacological Insights of Anisomeles indica Kuntze

    Anisomeles indica (L.) Kuntze is an ethnomedicinally important plant that has long been used in traditional medicine to treat a variety of ailments, including dyspepsia, abdominal pain, colic, allergies, inflammation, and rheumatic arthritis. However, the scientific framework underlying these medicinal properties is not well known. This study aimed to investigate the antidepressive, antidiarrheal, thrombolytic, and anti-inflammatory potential of a methanol extract of A. indica (MeOH-AI). The potential bioactive compounds in the MeOH-AI were identified using gas chromatography-mass spectrometry (GC-MS), and antidepressant activities were evaluated using the tail suspension test (TST) and forced swim test (FST). Antidiarrheal effects were also assayed in castor oil-induced diarrhea and gastrointestinal motility studies. The anti-inflammatory activities were explored by examining the effects on protein inhibition and denaturation in heat- and hypotonic solution-induced hemolysis assays. The thrombolytic activity was evaluated using the clot lysis test in human blood. BIOVIA and Schrödinger Maestro (v11.1) were applied for docking analysis to determine binding interactions, and the absorption, distribution, metabolisms, excretion/toxicity (ADME/T) properties of bioactive compounds were explored using a web-based method. The GC-MS analysis of MeOH-AI revealed the presence of several bioactive compounds. MeOH-AI administration resulted in significant (p < 0.01) reductions in the immobility times for both the FST and TST compared with those in the control group. MeOH-AI also induced significant (p < 0.01) reductions in castor oil-induced diarrhea severity and gastrointestinal motility in a mouse model. In addition, the in vitro anti-inflammatory and thrombolytic activity studies produced remarkable responses. The binding assay showed that 4-dehydroxy-N-(4,5-methylenedioxy-2-nitrobenzylidene) tyramine interacts favorably with monoamine oxidase and serotonin and M3 muscarinic acetylcholine receptors, displaying good pharmacokinetic properties, which may mediate the effects of MeOH-AI on depression and diarrhea. Overall, the research findings indicated that MeOH-AI has significant antidepressant, antidiarrheal, and anti-inflammatory effects and may represent an alternative source of novel therapeutic factors.

    Comparative valorisation on biological activities of Leea indica, Oroxylum indicum, Mikania micranth and Smilax macrophylla as a sustainable source of bioactive ingredients

    In Bangladesh, Leea indica, Oroxylum indicum, Mikania micrantha and Smilax macrophylla have been reported to have medicinal properties to treat numerous types of diseases, such as inflammation, pain, diarrhoea, skin problems and asthma. The present study designed in a comparative manner, assesses the qualitative phytochemical analysis, cytotoxic and antinociceptive activities of methanol leaves extract of L. indica (MELI), O. indicum (MEOI), M. micrantha (MEMM), and methanol extract of S. macrophylla leaves (MESMl) and stems (MESMs). The methanol extract of each plant comprises a significant quantity of secondary metabolites in the phytochemical study. At the same time, a non-significant weak to high toxicity in the brine shrimp lethality bioassay was assessed. Acetic acid and formalin-induced antinociceptive activity demonstrated highly significant (p < 0.001) dose-dependent pain reduction at a higher dose (400 mg/kg). Hence, current findings suggest that all five methanol extracts have potential antinociception effects along with weak to high cytotoxicity. Remarkably, these plants can be a successful substitute besides agri-food substances in the treatment of nociception.

    Investigation of the Pharmacological Properties of Lepidagathis hyalina Nees through Experimental Approaches

    Lepidagathis hyalina Nees is used locally in Ayurvedic medicine to treat coughs and cardiovascular diseases. This study explored its pharmacological potential through in vivo and in vitro approaches for the metabolites extracted (methanolic) from the stems of L. hyalina. A qualitative phytochemical analysis revealed the presence of numerous secondary metabolites. The methanol extract of L. hyalina stems (MELHS) showed a strong antioxidative activity in the 1,1-diphenyl-2- picrylhydrazyl (DPPH) and reducing power assays, and in the quantitative (phenolic and flavonoid) assay. Clot lysis and brine shrimp lethality bioassays were applied to investigate the thrombolytic and cytotoxic activities, respectively. MELHS exhibited an expressive percentage of clot lysis (33.98%) with a moderately toxic (115.11 g/mL) effect. The in vivo anxiolytic activity was studied by an elevated plus maze test, whereas the antidepressant activity was examined by a tail suspension test and forced swimming test. During the anxiolytic evaluation, MELHS exhibited a significant dosedependent reduction of anxiety, in which the 400 mg/kg dose of the extract showed 78.77 4.42% time spent in the open arm in the elevated plus maze test. In addition, MELHS demonstrated dosedependent and significant activities in the tail suspension test and forced swimming test, whereas the 400 mg/kg dose of the extract showed 87.67 6.40% and 83.33 6.39% inhibition of immobile time, respectively. Therefore, the current study suggests that L. hyalina could be a potential source of anti-oxidative, cytotoxic, thrombolytic, anxiolytic, and antidepressant agents. Further study is needed to determine the mechanism behind the bioactivities.

    Purchasing Power Parity in Developing - 8 Countries: Evidence from Time Series Data

    Purpose: This study aims to empirically assess the rationality of Purchasing Power Parity (PPP) theory in Developing-8 (D-8) countries. For this purpose, the data of PPP, Consumer Price Index (CPI), and Wholesale Price Index (WPI) were used from 1990 to 2015, with annual frequency. Methodology: This paper attempted to set up the long-run association between the nominal exchange rate and relative prices as opposed to old investigations. This study applies the ADF, PP, and the recently developed KPSS test to test data stationarity, followed by a cointegration test, granger casualty test, and vector error correction model. Findings: Time-series properties of this study specify that the real exchange rates are stationary for sample countries, indicating PPP holds in these countries, whereas Cointegration results demonstrate that a strong cointegrating relationship exists among the variables of Bangladesh and Turkey only. Practical Implications: The findings of the study have some policy implications, which suggest some recommendations for bilateral trade among these countries. Originality: Findings of the paper suggest that the structure of these developing eight (D-8) countries tends to be less diverse, and fewer economic changes occur than in developed countries. Limitations: Findings of this paper can vary on a different set of databases depending on the changing pattern of CPI and WPI
