K.Present study would be significant in the sustenance of quality characters for postharvest storage of Capsicum fruit with CO2-sensitization in biocompatible manner. The present experiment describes effects of CO2 sensitization on delaying postharvest ripening through physiological attributes in Capsicum fruit. The experiment was conducted with acidified bicarbonate-derived CO2 exposure for 2 h on Capsicum fruit, kept under white light at 25 °C through 7 days postharvest storage. Initially, fruits responded well to CO2 as recorded sustenance of greenness and integrity of fruit coat resolved through scanning electron micrograph. Loss of water and accumulation of total soluble solids were marginally increased on CO2-sensitized fruit as compared to non-sensitized (control) fruit. The ethylene metabolism biosynthetic genes like CaACC synthase, CaACC oxidase were downregulated on CO2-sensitization. Accompanying ethylene metabolism cellular respiration was downregulated on CO2 induction as compared to control through 7 days of storage. Fruit coat photosynthesis decarboxylating reaction by NADP malic enzyme was upregulated to maintain the reduced carbon accumulation as recorded on 7 days of storage under the same condition. CO2-sensitization effectively reduced the lipid peroxides as oxidative stress products on ripening throughout the storage. Anti-oxidation reaction essentially downregulates the ROS-induced damages of biomolecules that otherwise are highly required for food preservation during postharvest storage. Thus, the major finding is that CO2-sensitization maintains a higher ratio of unsaturated to saturated fatty acids in fruit coat during storage. Tissuespecific downregulation of ROS also maintained the nuclear stability under CO2 exposure. Cd contamination poses threats to plant growth as well as jeopardizing food safety and human health through the accumulation of Cd in edible parts of plants. Unraveling the Cd toxicity mechanisms and responses of plants to Cd stress is critical for promoting plant growth and ensuring food safety in Cd-contaminated soils. Toxicological research on plant responses to heavy metal stress has extensively studied Cd, as it can disrupt multiple physiological processes. In addition to morpho-anatomical, hormonal, and biochemical responses, plants rapidly initiate transcriptional modifications to combat Cd stress-induced oxidative and genotoxic damage. Various families of transcription factors play crucial roles in triggering such responses. Moreover, epigenetic modifications have been identified as essential players in maintaining plant genome stability under genotoxic stress. Plants have developed several detoxification strategies to mitigate Cd-induced toxicity, such as cell-wall binding, complexation, vacuolar sequestration, efflux, and translocation. This review provides a comprehensive update on understanding of molecular mechanisms involved in Cd uptake, transportation, and detoxification, with a particular emphasis on the signaling pathways that involve transcriptional and epigenetic responses in plants. This review highlights the innovative strategies for enhancing Cd tolerance and explores their potential application in various crops. Furthermore, this review offers strategies for increasing Cd tolerance and limiting Cd bioavailability in edible parts of plants, thereby improving the safety of food crops.Item Modulating reactive oxygen species and ion homeostasis for combined salt and cadmium stress tolerance in Brassica campestris: The role of beneficial microbes(Plant Stress, 2024-09-24) Hasanuzzaman, Mirza; Alam, Md. VaraThe land areas and crop species adversely impacted by salinity and heavy metals are growing rapidly. Current research indicates that plant growth-promoting microorganisms offer an environmentally friendly option for improving physiological and biochemical processes in plants growing under stress conditions. The aim of the present study was to investigate the potential mitigation of simultaneous salinity and cadmium (Cd) stress in rapeseed (Brassica campestris cv. BARI Sarisha-17) by the application of pirillum sp. (Az), phosphate solubilizing bacteria (PSB), potassium mobilizing bacteria (KMB), and vesicular arbuscular mycorrhiza (VAM). Seeds were treated with PSB or KMB prior to sowing, whereas Az, PSB, KMB, or VAM were added as supplements during soil preparation. At 21 days after sowing, the plants were treated with a combination of salt (100 mM NaCl) and Cd (0.25 mM CdCl ), with several applications at 7-day intervals. The combination of salt and Cd stress decreased plant growth and biomass, relative water content, and photosynthetic pigment levels, while also increased electrolyte leakage, lipid peroxidation, and the generation of excess reactive oxygen species (ROS). Salt and Cd stress also impaired plant ion balances of sodium, potassium and nitrate, antioxidant defenses, and glyoxalase system activity. Application of Az, PSB, or KMB restored these parameters to unstressed levels by facilitating the scavenging of ROS, maintaining water status, restoring ion balances, enhancing plant antioxidant defenses, and increasing glyoxalase enzyme activity, while reducing methylglyoxal toxicity and improving photosynthetic activity. The application of KMB was the most effective; however, all microbe supplementations showed the ability to alleviate the damage caused by stress in rapeseed. These findings highlight the ability of soil microorganisms with plant growth-promoting properties to improve the physiological and biochemical functions of rapeseed under Cd and salt stress.Item Multi-Trait Index-Based Selection of Drought Tolerant Wheat: Physiological and Biochemical Profiling(Plnts, MDPI, 2024-12-26) Mohi-Ud-Din, Mohammed; Hossain, Alamgir; Rohman, Motiar; Uddin, Nesar; Haque, Sabibul; Tahery, Mahmudul Hasan; Hasanuzzaman, MirzaDrought is a detrimental abiotic stress that severely limits wheat growth and productivity worldwide by altering several physiological processes. Thus, understanding the mechanisms of drought tolerance is essential for the selection of drought-resilient features and drought-tolerant cultivars for wheat breeding programs. This exploratory study evaluated 14 wheat genotypes (13 relatively tolerant, one susceptible) for drought endurance based on flag leaf physiological and biochemical traits during the critical grain-filling stage in the field conditions. Measurements included six physiological, seven gas exchange, six photosystem II, six stomatal, three reactive species, seven metabolomic solutes, and two biomass traits. All parameters were significantly influenced by drought, with varying genotypic responses. Hierarchical cluster analysis (HCA) categorized genotypes into three drought tolerance groups based on trait performance. Seven genotypes in Cluster 2 (BARI Gom 26, BARI Gom 33, BD-631, BD-600, BD-9910, BD-9889, BD-637) exhibited superior drought tolerance, characterized by minimal changes in physiological traits and biomass accumulation, reduced oxidative stress markers, and increased accumulation of osmoprotectants. The innovative multi-trait genotype-ideotype distance index (MGIDI) further ranked wheat genotypes in regard to drought tolerance, identifying BARI Gom 33, BARI Gom 26, BD-9889, and BD-600 as top performers. Notably, all these top-ranking genotypes belonged to Cluster 2, previously identified as the highest-performing group in the HCA. The identified genotypes with superior drought tolerance offer valuable genetic resources for enhancing wheat productivity in water-limiting environments. Traits related to photosynthetic activity, biomass gain, leaf conductance, water stress, and osmoprotection showed high selection differentials and heritability in MGIDI analysis, indicating their potential as selection targets for drought-tolerant wheat. Mitch(International Journal of Phytoremediation, 2024-10) Dolui, Debabrata; Das, Abir; Hasanuzzaman, Mirza; Adak, Malay KumarMethylene blue, a cationic dye as a pollutant is discharged from industrial effluent into aquatic bodies. The dye is biomagnified through the food chain and is detrimental to the sustainability of aquatic flora. Despite of number of physico-chemical techniques of dye removal, the use of aquatic flora for bio-adsorption is encouraged. Thus, we used Salvinia molesta D. Mitch in bio-reduction of methylene blue on concentrations of 0, 10, 20, and 30 mg L-1 through 5 days with biosorption kinetics. The dye removal was concentration-dependent, maximized at 2 days with 30 mg L-1 which altered the relative growth rate (44%) of plants. Biosorption recorded 71% capacity at optimum pH (8.0), 24 h reducing major bond energies of amide, hydroxyl groups, etc. Bioaccumulation of dye changed potassium content (446%) under maximum dye concentration modifying tissues for dye sequestration. Reactive oxygen species were altered on dye reduction by oxidase (33%) with redox homeostasis by enzymes. Plants altered the metabolism with over accumulation of polyamines (51%), abscisic acids (448%), and phosphoenolpyruvate carboxylase (83%) on dye reduction. Thus, this study is rationalized with a sustainable approach where aquatic ecosystems can be decontaminated from dye toxicity with the exercise of bioresources like Salvinia molesta D. Mitch as herein.Item Role of organic amendments in improving the morphophysiology and soil quality of Setaria italica under salinity(Heliyon, 2024-09) Irin, Israt Jahan; Hasanuzzaman, MirzaSalinity negatively impacts soil fertility by impairing the development and physiological func-tions of foxtail millet plants. Organic amendments have emerged as a viable solution in the reclamation and management of salinity inflicted soils and improve the performance of crop. In this regard, a pot experiment was carried out to examine the effect of organic amendments (OAs) on soil quality and its influence on the growth and physiology of foxtail millet under saline and non-saline condition. The findings indicated that under salt stress conditions, the levels of proline, hydrogen peroxide (H2O2), and electrolyte leakage (EL) risen, whilst other physiological pa-rameters decrease in foxtail millet. Under ideal conditions, they exhibit a rapid growth rate and can absorb a substantial amount of nutrients, macromolecules, and pollutants from bodies of water. Zinc oxide nanoparticles (ZnO NPs) synthesized from plant extracts, particularly under stress conditions, have opened new research avenues in the field of nanotechnology. Under salinity stress, the accumulation of flavonoids in duckweeds can affect the structure of ZnO NPs, helping researchers ascertain their antimicrobial role. In our study, we exposed mid-log phase duckweed monocultures to 75 mM NaCl in a full-strength Murashige and Skoog medium for 7 days, followed by a 15-day recovery period. We observed significant overexpression of superoxide and hydrogen peroxide as reactive oxygen species. As a result, chlorophyll and certain metabolites were produced in lesser amounts, while flavonoid and phenol content increased by 12% and 8%, respectively. This overproduction persisted up to 10 days into the recovery treatment period but dropped by 8% and 5%, respectively, by the 15th day. The flavonoid coating transformed the NPs into rosette clusters, which exhibited reduced antimicrobial activity against Aeromonas hydrophila, a Gram-negative, fish-pathogenic bacterium. Herein, we discuss potential mechanisms for the conformational transformation of ZnO NPs into finer dimensions in response to NaCl-induced oxidative stress in duckweed. In this study, the azo dye degradation capacity of salinity-treated plants increased as the flavonoid profile became enriched. Zinc oxide nanoparticles, both prior to and after salinity treatment, were found to be efficient in scavenging azo dye and mitigating its toxicity, as evidenced by improved germination, growth, and overall plant morphometry.Item Selenium and its nanoparticles modulate the metabolism of reactive oxygen species and morpho-physiology of wheat (Triticum aestivum L.) to combat oxidative stress under water deficit conditions(BMC Plant Biology, 2024) Hasanuzzaman, Mirza; Raihan, Md. VaraAbstract Background Wheat (Triticum aestivum L.) is one of the most important cereal crop species worldwide, but its growth and development are adversely influenced by drought stress. However, the application of trace elements is known to improve plant physiology under water-limited conditions. In this study, the effects of drought stress on wheat plants were investigated, with a focus on potential mitigation by foliar application of selenium nanoparticles (Se(np)) and sodium selenate (Na2SeO4). The experiment was conducted in a net house using a completely randomized design with four replications. The treatments involved three levels of drought stress (mild, moderate, and severe) started at 30 days after sowing (DAS), with foliar sprays of Se(np) and Se (both 25 µM) initiated at 27 DAS and repeated 4 times at 7-day intervals until 55 DAS. Results Drought stress significantly reduced plant growth, whereas Se(np) and Se sprays enhanced it. Drought stress induced chlorophyll degradation, increased malondialdehyde and hydrogen peroxide levels, impaired membrane stability, and caused electrolyte leakage. Severe drought stress reduced the levels of antioxidants (e.g., proline, ascorbate, and glutathione by 4.18-fold, 80%, and 45%) and the activities of antioxidant enzymes (ascorbate peroxidase, dehydroascorbate reductase, and others). Conversely, treatment with Se(np) and Se restored these parameters, for example, 1.23-fold higher total chlorophyll content with Se(np) treatment, 26% higher APX activity with Se treatment, 15% lower electrolyte leakage with Se treatment in wheat plants under severe drought stress. This study demonstrated the mitigation of the adverse effects of Li stress on tomato (Solanum lycopersicum L.) by the application of waste materials, namely coconut shell biochar (CBC) and steel slag (SS). To explore the impact of Li treatment on tomato plants different morphological, biochemical parameters and plant defense system were analyzed. Tomato plants exposed to Li had shorter roots and shoots, lower biomass and relative water contents, and showed decreases in physiological variables, as well as increases in electrolyte leakage and lipid peroxidation. However, the application of CBC and SS as passivators, either singly or in combination, increased growth variables of tomato and relieved Li-induced oxidative stress responses. The combined CBC and SS amendments reduced Li accumulation 82 and 90% in tomato roots and shoots, respectively, thereby minimizing the negative impacts of Li. Antioxidant enzymes SOD, CAT, APX and GR reflected 4, 5, 30, and 52% and glyoxalase enzymes I and II 7 and 250% enhancement in presence of both CBC and SS in Li treated soil, with a concurrent decrease in methylglyoxal content. Lithium treatment triggered oxidative stress, increased enzymatic and non-enzymatic antioxidant levels, and induced the synthesis of thiols and phytochelatins in roots and shoots. Hence, co-amendment with CBC and SS protected tomato plants from Li-induced oxidative damage by increasing antioxidant defenses and glyoxalase system activity. Both CBC, generated from agricultural waste, and SS, an industrial waste, are environmentally benign, safe, economical, and non-hazardous materials that can be easily applied on a large scale for crop production in Li-polluted soils. The present findings highlight the novel reutilization of waste materials as renewable assets to overcome soil Li problems and emphasize the conversion of waste into wealth and its potential for practical applications.Item Vanillic Acid Modulates Antioxidant Defense and Methylglyoxal Detoxification Systems to Combat Drought Stress in Tomato Seedlings(Plants, 2024-11) Parvin, Khursheda; Hasanuzzaman, Mirza; Mohsin, Sayed Mohammad; Nahar, KamrunVanillic acid (VA) regulates various plant physiological and biochemical processes upon different environmental stresses to enhance their tolerance. This study aimed to evaluate the protective effect of VA on growth and physiology, including osmoprotection, and antioxidant defense systems for enhancing higher tolerance by lowering oxidative damage against water deficit stress in tomatoes (Solanum lycopersicum L. cv. BARI Tomato-16). Hydroponically grown tomato seedlings (8 d old) were pretreated with 50 µM VA for 2 days followed by water deficit stress (imposed by water withdrawal and 12% polyethylene glycol; PEG-6000) for 4 d. Drought stress inhibited the seedlings' growth by reducing water content and photosynthetic pigments contents, alleviating oxidative stress induced by a reactive oxygen species and methylglyoxal. A significant enhancement in growth, biomass accumulation, and photosynthetic pigment content was observed in VA-pretreated stress conditions. In addition, there was an improvement in the water status and proline content, along with modulated activities of the antioxidant responses, including both non-enzymatic and enzymatic components in leaves of VA-pretreated seedlings upon the water deficit. Vanillic acid significantly reduced the reactive oxygen species generation and decreased cellular membrane damage in drought-affected tomato seedlings. Methylglyoxal detoxification was ensured to a great extent in VA-pretreated stressed tomato seedlings by strengthening the glyoxalase enzymes' activities. Therefore, VA can be effective for protecting tomato seedlings by inducing a plant antioxidant defense and the methylglyoxal detoxification system and osmoregulation under drought stress.Item Variations in photoperiods and their impact on yield, photosynthesis and secondary metabolite production in basil microgreens(BMC Plant Biol ., 2024-07-26) Fayezizadeh, Mohammad Reza; Ansari, Naser Alemzadeh; Sourestani, Mohammad Mahmoodi; Hasanuzzaman, MirzaAbstract Background: The effects of different photoperiods on plant phytochemical synthesis can be improved by adjusting the daily light integral. Photoperiod is one of the most important environmental factors that control growth, plant's internal rhythm and the synthesis of secondary metabolites. Information about the appropriate standard in terms of photoperiod for growing basil microgreens as one of the most important medicinal plants is limited. In this study, the effects of five different photoperiods, 6 (6 h × 3 cycles), 8 (8 h × 2 cycles), 16, 18, and 24 h day- 1 on the yield, photosynthesis and synthesis of secondary metabolites of three cultivars and one genotype of basil microgreens in floating system were evaluated. The purpose of this research was to determine the feasibility of using permanent light in growing basil microgreens and to create the best balance between beneficial secondary metabolites and performance. Results: The results showed that the effects of photoperiod and cultivar on all investigated traits and their interaction on photosynthetic pigments, antioxidant capacity, total phenolic compounds, proline content and net photosynthesis rate were significantly different at the 1% level. The highest levels of vitamin C, flavonoids, anthocyanins, yield and antioxidant potential composite index (APCI) were obtained under the 24-h photoperiod. The highest antioxidant capacity was obtained for the Kapoor cultivar, and the highest total phenolic compound and proline contents were measured for the Ablagh genotype under a 24-h photoperiod. The highest yield (4.36 kg m- 2) and APCI (70.44) were obtained for the Ablagh genotype. The highest nitrate content was obtained with a photoperiod of 18 h for the Kapoor cultivar. The highest net photosynthesis rate was related to the Violeto cultivar under a 24-hour photoperiod (7.89 μmol CO2 m- 2 s- 1). Antioxidant capacity and flavonoids had a positive correlation with phenolic compounds and vitamin C. Yield had a positive correlation with antioxidant capacity, flavonoids, vitamin C, APCI, and proline. Conclusions: Under continuous light conditions, basil microgreens resistance to light stress by increasing the synthesis of secondary metabolites and the increase of these biochemical compounds made basil microgreens increase their performance along with the increase of these health-promoting compounds. The best balance between antioxidant compounds and performance was achieved in continuous red + blue light. Based on these results, the use of continuous artificial LED lighting, due to the increase in plant biochemical with antioxidant properties and yield, can be a suitable strategy for growing basil microgreens in floating systems.
