Item EFFECT OF PLANT GROWTH REGULATORS ON IN VITRO MULTIPLICATION OF POTATO(DEPARTMENT OF GENETICS AND PLANT BREEDING, SHER-E-BANGLA AGRICULTURAL UNIVERSITY, DHAKA, BANGLADESH, 2010-06) HASAN, MD. Dhaka. during the period from Januan, 2009 to December 2009 to siudy the effecl of plant growth regulators NAA. I AA. RAP & GA3) on in vitro multiplication oF potato through callus culture and to find out the regeneration potentiality of three lotalo varieties Dianmnt. Cardinal and Granola using stem segment as e'.plants. Ditkrent concentrations and combinations oINAA and RAP were used on MS media for callus initiation and proliferation. Different concentrations and combinations of BAP and ()A were used with MS media for shoot induction and different concentrations and combinations 01' IAA and GA were used 11w rooting. the performance of cv. Cardinal was better for callus initiation. Minimum number of days required for callus initiation was recorded in cv. Cardinal produced on MS media supplemented with 3.0 mgI NA,\ and 1.0 mg/I RAP. But the highest Fresh weight of callus was found in cv. Diamant with the same concentration and combination of hormones at 60 DAt. in ease of number of shoots/callus, it was found that maximum number of shoots per calltis produced on MS media supplemented with 1.0 mgI RAP and 0.1 mg/I GA3 in cv. DialTiant and (iranulla at 60 DAI. But in case of days required for shoot induction. cv. Cardinal required minimum number of days. Itighest shoot length found in cv. Diamant with same combinations and concentrations of hormones at 60 DAt. Highest number of leaves per plantlets found in cv. Diamant and Cardinal produced on MS media supplemented with 1.0 mgI RAP and 0.1 mg1l GA3 at 60 DAt and niiniintim number ni leaves per plantlets found in cv. Granul Ia produced on MS media without any hormone at 60 DA I. Maxinium number of days required for root formation was noticed in cv. Granulla with 1.00 mgI IAA and 0.1 mg/I GAI and minimum number of days required for root llnnation was noticed in cv. Cardinal with 1.0 mng:I IA,\ and 0.05 mg I (A. I tighesi number of roots found in cv. Diamant with 1.0 ing I FAA and 0.05 mgI GA3 and minimum number of rooLs found in cv. Diamant and Grantilla with 1.0 rng1t IAt and 0.0 mg/I 6A3 at 60 t)AI. I lighcst root length found in cv. t)iamant with 1.() mg I IA A and 0.05 mng/L GA; niinimtlm root length found in cv. Grariutla with the same concentration and combinations of the same hormones at 60 DA I. The plantlets with sufficient number of well developed roots were successfully transferred in small holes of plastic iray in growth chamber and subsequently to soil in open atmosphere. Regenerated planticis from cv. Diamant survived easily at high rate in growth chamber and in open atmosphere.Item IDENTIFICATION OF BACTERIA AND THEIR LOADS ISOLATED FROM DIFFERENT BRANDS OF ICE CREAM(HAJEE MOHAMMAD DANESH SCIENCE AND TECHNOLOGY UNIVERSITY, DINAJPUR., 2012-12) HASAN, MD. KAMRUL; Dr. Md. Mostafizer RahmanA study was conducted to identify the organisms in ice cream manufactured by three different plants namely Igloo, Polar and Kwality sold in retail stores in Dinajpur town, Bangladesh. The samples were analyzed to determine the hygienic status of the ice creams and also for the total viable count, presence of Gram positive and Gram negative pathogenic bacteria.Three cups ice cream samples each of three popular varieties including Igloo, Polar and Kwality were collected from different retail stores in Dinajpur city. The bacteriological analyses were done at the Microbiology Laboratory of Hajee Mohammad Danesh Science and Technology University, Dinajpur. During July to December -2012. Total viable count (TVC) was performed according to the American Public Health Association, using plate count agar medium for TVC and Eosine methylene blue (EMB) agar media for total E. coli count and Staphylococcus agar no. 110 for total staphylococcus count. The average TVC for Kwality ice cream was lower (8.53 x 103 CFU/ml) in comparison to Polar ice cream (1.39 x 104 CFU/ml). It was apparent from the present study that the bacterial load of the ice cream samples of the three manufacturers were within the acceptable limit of public health safety, since the count was well below the acceptable limit. It was found that the highest extent of microbial contamination and proliferation of viable bacteria occurred in Polar ice cream. The average E. coli counts obtained from the study was in Igloo 9.26 x 103 CFU/ml (log 4.0), in Polar 1.14 x 10° CFU/ml (log 3.0) and in Kwality 7.95 x 102 CFU/ml (log 2.9) . The presence of numbers of E. coli in Igloo ice cream was little bit high indicated the poor hygienic practices during manufacture, post process contamination and unsatisfactory transportation. Statistically the E. coli were more closely related to total viable counts than the staphylococcal counts. The average staphylococcal counts in the samples of Igloo, Polar and Kwality were 2.60 x 10°CFU/ml (log 3.4), 0.0 CFU/ml (log 0.0), and 0.0 CFU/ml (log 0.0), respectively. The results demonstrated that kwality ice creams are of the superior quality product in respect of sanitary condition.Item NUTRIENT AVAILABILITY AND BODY COMPOSITION OF ADULT GIRIBAZ PIGEON (Columba livia) AND MEAT YIELD CHARACTERISTICS OF THEIR SQUAB(Dept. of Poultry Science, 2014-12) HASAN, MD. KAMRULThree experiments were conducted to evaluate nutrient availability, body composition of semi scavenging Giribaz pigeon and meat yield of their squab. The average DM, CP, CF, EE, ash, NFE, Ca and Av. P in crop and gizzard contents of male and female pigeon were 716.75, 108.50, 50.55, 15.90, 49.60, 574.25, 0.54 and 0.22 g/kg, respectively. The ME contents was 14.20 MJ/kg DM. The average live weight was 297.30g for male pigeon and 267.70g for female pigeon. Dressing percentage, total meat, breast meat, head and gizzard were 62.60, 34.52, 23.63, 3.09 and 1.91% for male pigeon and 56.80, 28.22, 19.45, 4.24 and 2.63% for female pigeon respectively. Live weight, dressing percentage, total meat and breast meat were (11.06, 5.80, 6.30 and 4.18%) higher (P<0.01) in males than females. While, females had heavier (P<0.01) head and gizzard (1.15 and 0.72 %) than males. Total meat, breast meat and wing meat were increased (P<0.01) with advancement of age of squab. Proportionate bone weight had no relation (P>0.05) with age. On the other hand, dressing percentage, total meat and breast meat were increased (P<0.01) with the increasing live weight. Weight of drumstick meat and total bone (neck weight+ wing bone+ vertebral column weight+ thigh bone + drumstick bone) did not maintain any relation (P<0.05) with live weight. It is concluded that feeds in crop and gizzard of pigeon were low in CP, EE, NFE, Ash, Ca and Av. P but higher in CF and ME. Live weight had pronounced effects on dressing percentage, total meat and breast meat in both adult and squab pigeons. Supplemental diet like CP, EE, NFE, Ash, Ca and Av. P were rich, whereas low in CF that could increase the production of semi scavenging pigeon. Nutrient availability and body composition between male and female pigeon is apparently negligible difference.Item SCREENING OF OKRA GERMPLASM BASED ON MORPHOLOGICAL AND PHYSIOLOGICAL CHARACTERISTICS(DEPARTMENT OF AGRICULTURAL BOTANY SHER-E-BANGLA AGRICULTURAL UNIVERSITY DHAKA-1207, 2015-06) HASAN, MD. KAMRULA field experiment was conducted at the research farm of Sher-e-Bangla Agricultural University, Dhaka-1207, from March to July, 2014 to evaluate promising cultivars of okra. Sixteen okra cultivars viz. Shaymol Bangla, Sarosh-3, BARI Dherosh-1, Porosh Plus, OK-285, Deb-412, Parvani Kranti, Kalatia, Green soft, Arka Anamika, Malvika, Green valley, Toa DK-2, Shehzadi, Gunjon, Kochi were used as treatments. The experiment was laid out in a Randomized Complete Block Design (RCBD) with three replications. Screening was done based on morphological features, yield and yield contributing features and physiological features. Leaves per plant were the maximum for Parvani kranti at harvest. Anthesis occurred in OK-285 prior to any other varieties. Minimum incidence of YVMV was found from the variety Sharosh-3. In all other morphological attributes viz. plant height, days required for 80% germination, pods plant-1 and pod yield as well as physiological attributes like net assimilation rate, intercellular CO2 concentration, respiration rate and stomatal conductance were the highest for variety Green valley. The highest pod yield (14.26 t/ha) was obtained from the variety Green valley which was closely followed by Porosh plus (10.22 t/ha) and the lowest yield was obtained from the variety Parvani kranti (5.23 t/ha). So, Green Valley was the best variety in respect of morphological and physiological characteristics among the studied varieties.Item SELECTION OF ELITE SOYBEAN (Glycine max L. Merrill) GENOTYPES BASED ON MORPHO-AGRONOMIC TRAITS(DEPARTMENT OF GENETICS AND PLANT BREEDING, 2020) HASAN, MD. KAMRULThe present experiment was conducted by using eighteen advanced genotypes of Soybean (Glycine max L. Merrill) for selecting most promising genotypes at the experimental field of Sher-e-Bangla Agricultural University (SAU), Sher-e- Bangla Nagar, Dhaka-1207 during the period from mid-December, 2018 to mid- April, 2019 based on some morphological traits. The experiment consisted of one factor with three replications. The results indicated that the genotypes differed regarding all the characters studied. The phenotypic variance and coefficient of variation were higher than the genotypic variance and coefficient of variation in all the characters studied. Moderate to high heritability was observed for all characters except leaf length and number of primary branches per plant. High heritability coupled with high genetic advance in percent mean were observed for germination percentage, days to 50% flowering, plant height, number of pod per plant, number of seeds per plant and thousand seed weight. The characters germination percentage, days to 50% flowering, plant height, leaf length, number of pod per plant, number of seeds per pod and number of seeds per plant showed positive direct effect on yield. Therefore, importance had to be given for these characters in further breeding program to improve soybean yield. All the genotypes were grouped into four clusters having 5, 2, 6 and 5 genotypes, respectively. Cluster III comprised the maximum number (6) of genotypes followed by cluster I(5) and IV(5). The highest inter cluster distance (26.186) was observed between the cluster I and II. Likewise highest distant genotypes were G11(AGS-205) and G18 (BINA SOYBEAN 3) (1.362). Among the characters studied days to 50% flowering, number of primary branches per plant, number of seeds per pod, thousands seed weight were the important component characters having higher contribution to the genetic divergence of soybean. As a result, these traits could be emphasized during selection of parents for hybridization.
