Hence the present investigation has designed to appraise antidiabetic and antioxidant principle of the methanolic extract of Marsilea quadrifolia (MEMQ). Methods: Hypoglycemic effect was evaluated in alloxan induced diabetic rat. The oral administration of plant extract at a dose of 300 mg/kg body weight was given to fasting glucose loaded rat with regard to normal control during 1 hr. study period and in alloxan induced (110 mg/kg body weight i.p.) diabetic rat in comparison with reference drug Metformin Hydrochloride (100 mg/kg) during 3 days study period. The antioxidant potential of MEMQ was checked by qualitative method and quantitatively through DPPH(1,1- diphenyl-2-picryl-hydrazyl) scavenging assay at 517 nm. Total phenolic content, total antioxidant capacity and reducing power activity was also assayed. Results: Considerable drop in elevated blood glucose level was observed in the alloxan induced diabetic (p<0.05 & p<0.001) rat. At a dose of 300 mg/kg the extract showed glucose level reduction of 47.57% in alloxan induced rat while 44.38% was found for Metformin after 3 days. Antioxidant activity using DPPH was found to increase in a concentration dependent manner with an IC 50 value of 96.37 ± 3.62μg/ml higher than the standard one, IC 50 16.59±0.59μg/ml. Total phenolic content was found 165.75 ± 0.961 mg/g in GAE and the total antioxidant capacity was equivalents of ascorbic acid (224.90 ± 1.42 mg/g). Conclusions: In all ways the extract showed significant antidiabetic and antioxidative potency. Ekramul; Islam, AnwarulIn course of investigation on natural antioxidants, the present study was aimed to report the antioxidant activities, both in vitro and in vivo, of the crude methanolic extracts of the whole plant of Oxalis corniculata Linn along with its various organic fractions. The different assay methods, including total antioxidant activity, scavenging free radical, authentic peroxynitrite, nitric oxide and reducing power assessment were used to evaluate the antioxidant potential of the crude extract and its organic fractions. The ethylacetate (EtOAc) fraction, showed strong activity in all the model systems tested and in peroxynitrite model this fraction (IC50 value of 2.29±0.18 μg mL-1) exerted three-fold stronger activity than standard penicillamine (IC50 value of 6.20±0.32 μg mL-1). The reducing power of the extract was found to be concentration dependent. The administration of the extract/fractions at a dose of 250 and 500 mg kg-1 body weight to the male Wistar rats increased the percentage inhibition of reduced glutathione, superoxide dismutase and catalase significantly. Whereas, lipid peroxidation level in hepatotoxic rats markedly decreased at a dose of 500 mg kg-1 body weight after 7 days. The total phenol and flavonoid content were also measured in the crude extract along with its organic fractions. The Brine shrimp lethality bioassay was used to determine the toxicity of the extracts and Vincristin sulphate was used as positive control. The dichloromethane (CH2Cl2) fraction showed highest activity (LC50 value of 29.02±1.16 μg mL-1) and other showed activity in the order of: EtOAc fraction >n-BuOH fraction > MeOH extract > aqueous fraction. Taken together, these results suggest that O. corniculata extract has strong antioxidant properties and further validate the traditional use of this plant.Item In vitro antioxidant and anti-neoplastic activities of Ocimum sanctum leaves in Ehrlich Ascites Carcinoma bearing mice(© 2011 Academic Journals Inc., 2011) Islam, M. EkramulThe present study was designed to investigate the antioxidant and anti neoplastic activities of Ocimum sanctum Linn. (OS) leaves. Antioxidant potential of the OS extract was evaluated in vitro by DPPH (1, 1diphenyl-2-picrylhydrazyl) and NO (Nitric Oxide) scavenging assay and reducing power assay method. OS extract showed prominent scavenging activity in all the methods with IC50 value of 16.39±0.31 and 16.20±0.33 μg mL-1 for DPPH and NO assay method, respectively. In reducing power assay, OS extract also showed significant (p<0.001) activity. In addition, total phenolic and flavonoid content and total antioxidant capacity were also determined. The anti neoplastic effect of the leaves of OS against Ehrlich Ascites Carcinoma (EAC) in mice at the doses of 50 mg kg-1 body weight intraperit one ally. Significant (p<0.001) increases of survival times 33±1.81 days for crude extract of the (50 mg kg-1) treated tumor bearing mice were confirmed with respect to the control group (20±0.12 days). Hematological studies reveal that the Heamoglobin (Hb) content was decreased in EAC treated mice whereas restoration to near normal levels was observed in extract treated animals. There was a significant (p<0.001) decrease in RBC count and increase in WBC counts in extract/fraction treated animals when compared to EAC treated animals. From the result it was showed that the extract has significant antioxidant as well as anti neoplastic activity.
