    CHARACTERIZATION OF BACTERIAL PATHOGENS FROM CALF DIARRHOEA WITH ANTIBIOGRAM STUDY OF IDENTIFIED FIELD ISOLATES

    Calf diarrhosea is a common problem in among small, medium and large cattle farms of Bangladesh. High economic loss occurs due to calf diarrhea. Indiscriminate use of antibiotic to control the disease results drug resistance and limits the therapeutic possibilities in the treatment of the disease. The research work was conducted to Isolate, Identify and characterize the bacterial pathogens causing calf diarhoea. The study was conducted on 114 faecal samples collected directly from the rectum of diarrhoeic calves’ dairy farm and brought for the treatment in the District and Upazilla hospitals under Panchagarh District to find out the occurrence of pathogenic bacteria. Out of 114 fecal samples exmined for bacterial isolates, the number and frequency of distribution in percentage for positive samples were 84 (73.7%) of them for E.coli 44(38.6%), Salmonella spp 25(21.9%), Staphylococcous 15(13.2%), other bacteria 18(15.8%) and 12 (10.5%) was negative for bacteria (Nutritional and other than bacterial causes involved).The frequency response of E.coli , Salmonella and Staphylococcous to Microscopic examination and specific biochemical tests were 58.6%, 31.3% and 10.01% respectively. Isolation and identification of the microorganisms were confirmed on the basis of their morphology, staining, cultural and biochemical properties. Antibiogram of the isolated organisms was also studied. All the E.coli isolates were found to produce bright pink colonies on MacConkey agar and characteristic yellowish green metallic sheen colonies on EMB agar. Salmonella produce opaque, translucent and colorless colonies on SS agar but on BGA it produces pale, pink color colonies against pinkih background. Staphyloccous produce growth of circular, small, smooth, convex and golden yellowish colonies on Nutrient agar, produce hemolysis on blood agar media and Staphylococcous agar no.10 media produce yellowish colonies. The Staphyloccous were produced acid. The change of color from reddish to yellow indicated acid production. In TSI agar slant, yellow slant, yellow butt, absence of gas bubbles and absence of black precipitate, The results of MR test positive, V- P test negative, Indole test negative.The results of MR and Indole tests of E.coli isolates were positive; VP test was negative. In case of Salmonella MR Test was positive but VP and Indole test were negative. The antibiogram study revealed that these three isolates were highly sensitive to Ciprofloxacin, Azithromycin, Gentamicin, moderately sensitive to Chloramphenicol, Enrofloxacin and resistant to Amoxycilli, Ampcillin, Penicillin and Bactericin. The findinings of the experiment indicates that the use of Ciprofloxacin, Azithromycin and Gentamicin may have the preference to be choice in clinical control of Salmonella, E.coli, and Staphyloccous causing calf diarrohoea in Bangladesh.

    EFFECTS OF POTASSIUM ON GROWTH, YIELD AND QUALITY CHARACTERS IN TOMATO

    The growth, yield contributing characters and yield of tomato largely depend on soil and climatic conditions and also on variety. Among these, proper doses of potassium and variety play a vital role. The field experiment was conducted at Sher-e-Bangla Agricultural University Farm, Dhaka, Bangladesh during October 2015 to April, 2016 to evaluate the effect of potassium on yield and quality of tomato. Two different varieties, viz., BARI Tomato-2, Minto hybrid Tomato and five doses of potassium viz. P0=0 kg/ha, P1= 140 kg/ha, P2= 150 kg/ha, P3= 160 kg/ha, P4= 170 kg/ha were used to conduct this experiment. The experiment was laid out in Randomized complete Block Design (RCBD) having two factors and replicated three times. Data were collected on plant height, number of branches per plant, length of branch, number flowers cluster per pant, number of fruit per plant, yield per plot, yield per hectare, TSS, β- Carotene content and Vit C content. A statistically significant variation was recorded in terms of all the characters related to growth and yield of tomato. The maximum plant height, number of branches per plant, length of branch was produced by Minto hybrid. The maximum number flowers cluster per plant and number of fruit per plant was produced by Minto hybrid. The maximum yield of fruits per hectare (41.60 tones) was obtained from Minto hybrid. The maximum plant height, number of branches per plant, length of branch was observed in doses of potassium 160 kg/ha. The maximum number flowers cluster per plant and number of fruit per plant was produced by doses of potassium 160 kg/ha. The maximum yield of fruits per hectare (35.24 tones) was obtained from doses of potassium 160 kg/ha treatment. The highest yield of fruits per hectare (49.01 tones) was obtained from Mintro hybrid Tomato with 160 kg K/ha. The lowest yield of fruits per hectare (12.22 tones) was obtained from BARI Tomato-2 with 0 kg K/ha.

    INFLUENCE OF GA3 AND NAA ON GROWTH, YIELD AND QUALITY OF OKRA (Abelmoschus esculentus L.)

    A field experiment was carried out to assess the performance of okra cv. Green Finger as influenced by GA3 and NAA during the period from February 2023 to June, 2023 at area sales center (ASC), Bangladesh Agricultural Development Corporation (BADC), Chehelgazi, Dinajpur, Bangladesh. The treatments consisted of three levels of GA3 (i.e., 0 ppm, 150 ppm and 250 ppm) combined with three levels of NAA (i.e., 0 ppm, 150 ppm and 250 ppm) and formed a total of nine treatment combinations. All the treatments were replicated thrice in the two factorial Randomized Complete Block Design (RCBD). The results revealed that plant growth and yield as well as qualitative attributes of okra were highly influenced by different levels of GA3, NAA both individually or in combination. Vegetative growth like plant height, number of leaves and branch plant-1 at 20, 40, 60 and 80 DAS were statistically higher than control. At 80 DAS, the maximum plant height (114.40 cm), leaves plant-1 (57.03), and branch plant-1 (5.43) were observed by NAA @ 150 ppm + GA3 @150 ppm. Further the leaves area (112.10 cm2), internode number (25.40), internode length (21.33 cm), buds plant-1 (51.13), pods plant-1 (24.63), pod length (15.93 cm), pod diameter (2.02 cm), dry matter of pod (21.33 %), 1000 seeds weight (75.80 g), stover yield plant 1(505.67 g), yield plant-1 (419.93 g) and yield plot-1 (14.43 ton) were also found to be superior by NAA @ 150 ppm + GA3 @ 150 ppm as compared to control. In addition, the maximum amount of β-carotene (69.46 mg), TSS (4.53 %) and vitamin C were found from NAA @ 150 ppm + GA3 @ 150 ppm respectively, and minimum (43.24 mg, 6.45 mg and 2.50 %) were observed from N0G0, N2G0 and N1G2, respectively. Moreover, minerals like magnesium and zinc were also increased and potassium was decreased by the application of different levels of GA3 and NAA as compared to the control whereas calcium was not influenced by GA3 and NAA. Overall, the present experiment suggested that the application of GA3 @ 150 ppm + NAA @ 150 ppm can be effective in enhancing the growth, yield and quality of okra.
